摘  要:目的研究人胎脑来源神经干细胞制剂在免疫缺陷小鼠中的致瘤性,探讨直接移植神经干细胞临床试验研究的安全性和可行性。方法选取吉林大学通源生物工程有限公司提供的人胎脑神经干细胞(原代)为实验组,以DPBS为阴性对照组,以Hela细胞为阳性对照组,分别于各组免疫缺陷裸鼠的肋部皮下注射人胎脑神经干细胞、DPBS和Hela细胞,注射体积为0.2ml/只,含DPBS、神经干细胞2×10^(7)个或Hela细胞1×10^(6)个。每天观察小鼠一般生理指标(如运动行为、腺体分泌、粪便形态以及体表粘膜等),并于注射后第1、2、3、6个月对部分小鼠注射部位进行病理切片检查(HE染色),以确定是否有瘤变情况,并进行统计学分析。结果实验组和DPBS阴性对照组均无致瘤性,Hela细胞阳性对照组有致瘤性。结论直接移植人胎脑神经干细胞后无致瘤性,为其进入临床试验研究的安全性和可行性提供依据。Objective To study the tumorigenicity of neural stem cells derived from human fetal brain in immunodeficient mice,and to explore the safety and feasibility of direct transplantation of neural stem cells.Methods Human fetal brain neural stem cells(primary generation)from Jilin University Tongyuan Bioengineering Co.Ltd.were selected as experimental group,DPBS as negative control group,and Hela cells as positive control group,human fetal brain neural stem cells,DPBS and Hela cells was injected subcutaneously into the ribs of immunodeficient nude mice in each group.The injection volume was 0.2ml for each mice,containing DPBS,2×10^(7)neural stem cells or 1×10^(6) Hela cells.General physiological indicators of mice,such as exercise behavior,gland secretion,feces and body surface mucosa,were observed daily.Pathological examination(HE staining)was conducted in some mice injection site at 1,2,3,6 months after injection,determined whether there was neoplasia with statistical analysis.Results There had no tumorigenicity in experimental group and DPBS negative control group,but Hela cells positive control group had tumorigenicity.Conclusion Direct transplantation of neural stem cells from human fetal brain has no tumorigenicity,which provides basis for its safety and feasibility in clinical trials.
