摘  要:目的:观察生地黄的抗焦虑作用,明确其抗焦虑作用部位并探讨作用机制。方法:复制小鼠焦虑模型、阴虚焦虑复合模型,观察小鼠在高架十字迷宫实验(elevated plus-maze,EPM)中开臂滞留时间百分比(percentage of open arm retention time,OT%)、开臂进入次数百分比(percentage of open arm entry,OE%)以及明暗箱实验中的穿梭次数以评价生地黄的抗焦虑药效;检测脑组织中γ-氨基丁酸(gamma aminobutyric acid,GABA)、谷氨酸(glutamate,GLU)、5-羟色胺(5-hydroxytryptamine,5-HT)的含量,测定阴虚焦虑小鼠血清中三碘甲状腺原氨酸(triiodothyronine,T_(3))、甲状腺素(thyroxine,T_(4))、促甲状腺激素(thyroid stimulating hormone,TSH)的含量以探讨生地黄抗焦虑的作用机制。结果:焦虑小鼠模型实验中,与焦虑模型组比较,生地黄及其多糖能显著增加焦虑小鼠的OT%、OE%和穿梭次数(P<0.05),显著升高焦虑小鼠脑组织中GABA的含量(P<0.05),降低GLU、5-HT的含量(P<0.05,P<0.01)。阴虚焦虑小鼠模型实验中,与阴虚焦虑模型组比较,生地黄及其多糖能显著增加阴虚焦虑小鼠的OT%、OE%和穿梭次数(P<0.05,P<0.01),显著升高脑组织中GABA的含量,降低GLU、5-HT的含量(P<0.05,P<0.01),并能显著降低阴虚焦虑小鼠血清中T_(3)、T_(4)、TSH的含量(P<0.05,P<0.01)。结论:生地黄具有抗焦虑作用,有效部位为生地黄多糖,其作用机制可能与升高小鼠脑组织中GABA的含量、降低GLU、5-HT的含量、抑制阴虚小鼠下丘脑-垂体-甲状腺(hypothalamus-pituitary-thyroid gland,HPT)轴亢进有关。Objective:To observe the anti-anxiety effect of Rehmannia glutinosa and explore its mechanism.Method:The anxiety model and Yin-deficiency anxiety complex model of mice were duplicated,and the percentage of open arm retention time(OT%),percentage of open arm entry(OE%)and shuttle times were observed in Elevated Plus Maze(EPM)test.The contents of GABA,Glu and 5-HT in brain tissue were detected,While T3,T4 and TSH in serum of Yin-deficiency anxiety complex model mice were tested.Results:In the anxiety model experiment,compared with the anxiety model group,Rehmannia glutinosa and Rehmannia glutinosa polysaccharide increased the shuttle times,OT%and OE%(P<0.05),increased the content of GABA significantly(P<0.05),and decreased the content of GLU and 5-HT(P<0.05,P<0.01)in brain tissue of the anxiety model mice.In the Yin-deficiency anxiety complex model experiment,compared with the Yin-deficiency anxiety complex model group,Rehmannia glutinosa and Rehmannia glutinosa polysaccharide could significantly increase OT%,OE%and shuttle times(P<0.05,P<0.01),increase GABA content,reduce GLU,5-HT content(P<0.05,P<0.01)in brain tissue and the contents of T_(3),T_(4) and TSH in serum of the yin-deficiency anxiety complex model significantly(P<0.05,P<0.01).Conclusion:Rehmannia glutinosa has anti-anxiety effect on anxiety mice and Yin-deficiency anxiety mice.The effective part is Rehmannia glutinosa polysaccharide.The mechanism of anti-anxiety effect may be related to increasing the content of GABA in brain tissue,reducing the content of GLU and 5-HT,and inhibiting the hyperactivity of HPT axis in Yin-deficiency mice.
