磷酸肌酸对糖尿病大鼠冠状动脉的保护研究

摘  要:目的:建立糖尿病大鼠心肌缺血再灌注(MIRI)模型,观察磷酸肌酸(PCr)对冠状动脉的保护作用。方法:建立糖尿病8周大鼠模型,随机分为4组(n=8):假手术组(Ⅰ)、MIRI+生理盐水组(Ⅱ)、MIRI+PCr单次注射组(Ⅲ)、MIRI+PCr多次注射组(Ⅳ),分离冠脉左前降支,观察冠脉对Ach和5-HT舒缩反应性的变化;电镜观察冠脉超微结构的变化。结果:冠脉对Ach的舒张反应,同一浓度下与Ⅰ组相比,Ⅱ组的舒张率下降(P<0.05),Ⅲ、Ⅳ组无统计学差异(P>0.05)。对5-HT的收缩反应,同一浓度下与Ⅰ组相比,Ⅱ、Ⅲ组收缩率明显升高(P<0.01),Ⅳ组无统计学差异(P>0.05);与Ⅱ、Ⅲ组相比,Ⅳ组收缩率明显下降(P<0.01)。与Ⅱ组相比,Ⅲ、Ⅳ组冠脉的病理损伤均减轻。结论:PCr对心肌缺血再灌注糖尿病大鼠冠状动脉具有保护作用。Objective:To investigate the protective effects of phosphocreatine(PCr)on the coronary artery following myocardial ischemia reperfusion injury(MIRI)in diabetic rats.Methods:The diabetic rats which were successfully induced for 8 weeks were randomly divided into four groups(n=8):sham-operation group(Ⅰ),MIRI+saline group(Ⅱ),MIRI+PCr administrated for single does group(Ⅲ),MIRI+PCr administrated for multiple does group(Ⅳ).End of the experiment,separating the LAD and making the artery ring,to observe the reactivity of coronary artery to Ach and 5-HT.Electron microscope was used to observe the ultrastructure of coronary artery.Results:The relaxation of coronary artery to Ach,at the same concentration,compared with groupⅠ,groupⅡwas significantly lower(P<0.05),groupⅢandⅣall had no statistically differences(P>0.05).While the contraction to 5-HT,at the same concentration,compared with groupⅠ,groupⅡandⅢsignificantly increased(P<0.01),groupⅣhad no statistically difference(P>0.05);compared with groupⅡandⅢ,groupⅣsignificantly decreased(P<0.01).The pathological damage to coronary artery in groupⅢandⅣwere significantly less than groupⅡ.Conclusion:PCr can protect coronary artery against MIRI injury in diabetic rats.
