摘　　要：目的检测circ_0000620在肺腺癌患者组织中的表达并且分析其与患者化疗耐药的相关性。方法选取2018年1月至2020年1月在郑州大学第一附属医院确诊的120例肺腺癌患者的癌组织及癌旁组织,其中男73例,女47例,年龄≥55岁者71例,<55岁者49例,化疗耐药者28例,化疗敏感者92例。采用环状RNA(circRNA)高通量测序技术筛选出差异性表达的circRNA;使用divergent primer和测序技术证明circ_0000620在肺腺癌组织中是否存在且为环状;采用实时荧光定量聚合酶链式反应(qRT-PCR)检测人肺腺癌组织、癌旁组织、肺腺癌细胞株(A549、PC-9、SPC-A-1、NCI-H1299、NCI-H1395)和正常人支气管上皮细胞(NHBE)中circ_0000620的表达情况。结果circ_0000620在肺腺癌组织中存在且为环状;肺腺癌组织中circ_0000620的表达水平高于其配对癌旁正常组织,差异有统计学意义(P<0.05);与化疗敏感组相比,化疗耐药组circ_0000620的表达水平升高,差异有统计学意义(P<0.05),而比较不同性别、不同年龄组中circ_0000620的表达水平,差异无统计学意义(P>0.05)。结论肺腺癌患者组织中circ_0000620高表达,且在患者化疗耐药组中水平更高。Objective To investigate the expression of circ_0000620 in lung adenocarcinoma tissues and analysis the relationship between circ_0000620 and chemotherapy resistance.Methods A total of 120 pairs of lung adenocarcinoma and paracancerous tissues in the First Affiliated Hospital of Zhengzhou University from January 2018 to January 2020 were selected,of which 73 were males,47 were females,71 cases were≥55 years old,49 cases were<55 years old,28 cases were chemotherapy resistant,and 92 cases were chemotherapy sensitive.High throughput sequencing of circular RNA(circRNA)was used to select the distinct expression of circRNA.The divergent primer and sequencing technology were used to confirm whether circ_0000620 exists in lung adenocarcinoma and was a circular structure.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the expression of circ_0000620 in lung adenocarcinoma tissue,paracancerous tissue,lung adenocarcinoma cell lines(A549、PC-9、SPC-A-1、NCI-H1299、NCI-H1395)and normal human bronchial epithelial cell(NHBE).Results It was proved that circ_0000620 existed in lung adenocarcinoma tissue and was a circular structure.Cire_0000620 showed high expression in lung adenocarcinoma tissue and that than its counterpart in normal tissue adjacent to carcinoma,the difference was statistically significantly(P<0.05).Compared with chemotherapeutic sensitive group,the expression level of circ_0000620 in chemotherapy resistant group was increased,and the difference was statistically significantly(P<0.05),while the expression level of circ_0000620 in different genders and age groups were not significantly different(P>0.05).Conclusion The expression level of circ_0000620 is significantly increased in lung adenocarcinoma and the level was higher in the chemotherapy resistant group.
