摘 要:目的探讨miR-145靶向调节ABCE1的分子机制及其在抗乳腺癌细胞增殖中的作用。方法利用生物信息学方法筛选出靶向调节ABCE1的miR-145,取乳腺癌组织和癌旁组织检测ABCE1和miR-145的表达水平。通过在乳腺癌细胞MCF-7中转染miR-145 mimic和inhibitor,分析miR-145对ABCE1的蛋白表达及细胞凋亡的影响。采用荧光素酶报告基因实验验证miR-145对ABCE1 mRNA的靶向调节作用。结果在乳腺癌组织中ABCE1蛋白的表达明显高于癌旁组织,同时miR-145在癌组织中的含量显著降低。在MCF-7细胞中转染miR-145 mimic后能抑制ABCE1的表达,同时促进细胞凋亡。此外,在293T细胞中共转染miR-145 mimic和psiCHECK-ABCE1-3'UTR后,发现荧光素酶活性受到明显抑制。结论miR-145可通过靶向沉默ABCE1 mRNA来抑制乳腺癌细胞增殖,提示miR-145对ABCE1的调控可作为治疗乳腺癌的途径之一。Objective To determine the molecular mechanism of miR-145 targeting and regulating ABCE1 and its role in the anti-proliferation of breast cancer cells.Methods Bioinformatics methods were used to screen out miR-145 that targeted and regulated ABCE1.Breast cancer tissues and adjacent tissues were taken to detect the expression levels of ABCE1 and miR-145.By transfecting miR-145 mimic and inhibitor into breast cancer cell MCF-7,the effect of miR-145 on ABCE1 protein expression and cell apoptosis was analyzed.The luciferase reporter gene experiment was used to verify the targeted regulation of miR-145 on ABCE1 mRNA.Results The expression of ABCE1 protein in the breast cancer tissues was significantly higher than that in the adjacent tissues,and the content of miR-145 in the cancer tissues was reduced.Transfection of miR-145 mimic in MCF-7 cells inhibited the expression of ABCE1 and promoted the cell apoptosis.In addition,after co-transfection of miR-145 mimic and psiCHECK-ABCE1-3'UTR in 293T cells,luciferase activity was significantly inhibited.Conclusion MiR-145 can inhibit the proliferation of breast cancer cells by targeting ABCE1 mRNA silence,suggesting that the regulation of ABCE1 by miR-145 can be used as a treatment for breast cancer.
