摘 要:目的:探究miR-320a靶向Pirh2对非小细胞肺癌(NSCLC)细胞增殖和迁移的调控作用。方法:采用qRT-PCR检测人NSCLC癌组织、癌旁组织和细胞株中miR-320a mRNA表达水平。采用miR-320a mimics质粒转染,CCK8法检测NSCLC细胞增殖能力,Transwell实验检测细胞迁移和侵袭能力,TargetScan生物信息学软件预测miR-320a与Pirh2的相互关系,并通过荧光素酶报告基因验证。采用siRNA干扰细胞内Pirh2表达,观察Pirh2基因沉默后细胞增殖、迁移和侵袭能力。通过共转染miR-320a mimics和pcDNA3.1-Pirh2质粒,观察Pirh2表达增加后对细胞增殖、迁移和侵袭能力的影响。结果:人NSCLC癌组织和细胞株中miR-320a表达水平降低。过表达miR-320a明显抑制NSCLC细胞增殖和迁移。TargetScan生物信息学软件预测显示miR-320a与Pirh2的3′UTR存在结合位点,荧光素酶报告实验显示miR-320a mimics显著降低WT-Pirh23′UTR的荧光素酶活性。Pirh2基因干扰后细胞增殖和迁移能力显著降低,共转染miR-320a mimics和pcDNA3.1-Pirh2质粒后,细胞增殖和迁移能力显著高于转染miR-320a mimics+pcDNA3.1细胞。结论:miR-320a通过下调Pirh2抑制NSCLC细胞增殖和迁移。Objective:To investigate the regulatory effect of miR-320a on the proliferation and migration of non-small cell lung cancer(NSCLC)cells by targeting ubiquitin ligase Pirh2.Method:qRT-PCR assay was used to detect the expression of miR-320a mRNA in the tumor tissues,para-carcinoma tissues,and NSCLC cell lines.The effect of miR-320a on the proliferation and invasion of NSCLC cells was analyzed by overexpression miR-320a and/or knockdown of Pirh2 in NSCLC cell lines.The targeting relationship between miR-320a and Pirh2 was confirmed by dual-luciferase reporter assay and Western blot detection.The proliferation and invasion abilities of cells were detected by CCK-8,colony formation assay,and Transwell assay,respectively.Results:The expression of miR-320a was decreased in NSCLC tissues and cells.Moreover,overexpression of miR-320a Pirh2 could inhibit the proliferation and invasion of NSCLC cells.The results of the dual-luciferase reporter assay showed that the Pirh2-3′UTR-WT luciferase activity of NSCLC cells in the miR-320a mimics group was decreased.Besides,the ability of cell proliferation and migration in the miR-320a mimics+pcDNA3.1-Pirh2 group was more than that in the miR-320a mimics+pcDNA3-1 group significantly.Conclusion:miR-320a can inhibit the proliferation and migration of NSCLC cells by downregulating Pirh2.
