摘　　要：目的:分析胃癌组织中单酰基甘油酰基转移酶3(MOGAT3)的表达情况及其与胃癌患者术后生存时间的关系。方法:利用GEPIA和Oncomine数据库信息分析MOGAT3 mRNA在胃癌组织中的表达情况,采用组织芯片结合免疫组织化学Envision法检测135例胃癌组织石蜡标本MOGAT3蛋白的表达,分析MOGAT3蛋白表达与患者临床病理参数及术后生存时间的关系。结果:数据库信息分析显示,胃癌组织中MOGAT3 mRNA表达高于癌旁正常胃组织,差异有统计学意义(P<0.01)。利用组织芯片技术联合免疫组化检测显示,胃癌组织中MOGAT3蛋白表达率为80.0%,显著高于癌旁组织的表达率31.9%,差异有统计学意义(P<0.001)。MOGAT3蛋白阳性表达与患者TNM分期、浸润程度、淋巴结转移及术前血CEA水平显著相关(P<0.05)。生存分析发现,MOGAT3蛋白阳性患者总生存率显著低于阴性患者,差异有统计学意义(P<0.05)。COX回归分析显示,MOGAT3蛋白高表达、TNM高分期、淋巴结转移是胃癌患者预后的独立危险因素。结论:MOGAT3蛋白可能促进胃癌的发生发展,有助于预测胃癌患者的预后,有望成为胃癌治疗的新靶点。Objective:To study the expression of MOGAT3 in gastric cancer and its relationship with the survival time of patients with gastric cancer.Methods:GEPIA and Oncomine were used to analyze the expression of MOGAT3 mRNA in gastric cancer.Tissue chip combined with immunohistochemical(IHC)Envision method was used to detect the expression of MOGAT3 protein in 135 cases of gastric cancer paraffin specimens。The relationship between the expression of MOGAT3 protein and clinical pathological parameters,overall survival time and the prognosis of patients after surgery were then analyzed.Results:The results obtained from the databases showed that MOGAT3 mRNA in gastric cancer tissues was higher than that in adjacent normal gastric tissues,and the difference was statistically significant(P<0.01).Tissue chip technology combined with IHC analysis showed that the expression rate of MOGAT3 protein in gastric cancer tissues was 80.0%,which was significantly higher than that in adjacent normal gastric tissues(31.9%),and the difference was statistically significant(P<0.001).MOGAT3 protein positive expression was significantly related to high TNM stage,deeper invasion(T),lymph node metastasis(N),and preoperative high CEA(P<0.05).Survival analysis showed that the overall survival rate of MOGAT3 protein positive expression patients was significantly lower than that of negative expression patients,and the difference was statistically significant(P<0.05).COX regression analysis showed that MOGAT3 protein high expression,high TNM stage,and lymph node metastasis were independent risk factors for the prognosis of gastric cancer patients.Conclusion:MOGAT3 protein may promote the occurrence and development of gastric cancer,and predict the prognosis of gastric cancer patients,which is expected to become a new target for gastric cancer treatment.
