摘 要:灵芝发酵过程中的营养条件可显著提高灵芝多糖的产量、抗氧化活性和抑菌活性。本研究以灵芝为主要材料,通过单因素实验探究发酵时培养条件pH、温度、转速、通气比和营养条件碳源、氮源对灵芝子实体生长情况及多糖产量的影响,再经过3因素优化得到灵芝深层发酵产多糖的最佳条件为:温度28℃,pH值6.0,转速150rpm,通气比为2,葡萄糖含量2%,蛋白胨含量0.4%,培养时间168h。该实验结果经过1t的中试实验得以验证,未经优化培养条件发酵浸膏多糖含量为3.6%~4.2%,优化后的多糖得率可达10.1%~10.6%,较未经优化培养条件的发酵多糖得率提高6%。The nutritional conditions during Ganoderma lucidum fermentation can significantly improve the yield,antioxidant activity and antibacterial activity of Ganoderma lucidum polysaccharide.In this study,Ganoderma lucidum was used as the main material to explore the effects of culture conditions,pH,temperature,rotating speed,aeration ratio and nutritional conditions,carbon source and nitrogen source on the growth of Ganoderma lucidum fruiting body and polysaccharide yield through single factor experiment.After three factors optimization,the optimal conditions for Ganoderma lucidum submerged fermentation to produce polysaccharide were as follows:temperature 28℃,pH 6.0,rotating speed 150 rpm,aeration ratio 2,The content of glucose was 2%,the content of peptone was 0.4%,and the culture time was 168h.The experimental results were verified by 1 ton pilot experiment.The polysaccharide content of fermentation extract without optimized culture conditions was 3.6%~4.2%,and the yield of polysaccharide after optimization was 10.1%~10.6%,which was 6%higher than that without optimized culture conditions.
