摘 要:目的观察“原络通经”针刺法对SAMP8小鼠学习记忆能力的影响,探究“原络通经”针刺法促进痴呆小鼠学习记忆能力恢复的作用机制。方法选取SAMR1小鼠6只作为SAMR1组,常规饲养,仅每天抓取;另选取SAMP8小鼠18只,采用随机数字表法分成SAMP8空白组、“原络通经”组和百会组各6只,SAMP8空白组仅每天抓取,“原络通经”组和百会组给予相应针刺干预。各组干预30 d后,采用Morris水迷宫实验检测小鼠学习记忆与空间探索能力,HE染色观察小鼠海马区神经细胞变化,Western blot法检测海马组织中N-甲基-D-天冬氨酸受体(NMDAR)表达情况,ELISA法检测小鼠海马组织中Aβ_(40)蛋白含量。结果与SAMP1组比较,SAMP8空白组小鼠逃避潜伏期明显延长,原平台象限停留时间明显缩短,穿过原平台位置次数明显减少,差异均有统计学意义(P均<0.05);与SAMP8空白组比较,“原络通经”组与百会组SAMP8小鼠逃避潜伏期均明显缩短,原平台象限停留时间明显延长,穿过原平台位置次数明显增加,且“原络通经”组改善情况均明显优于百会组,差异均有统计学意义(P均<0.05)。海马HE染色显示SAMR1组小鼠海马组织神经细胞形态、结构正常;SAMP8空白组小鼠海马组织神经细胞皱缩,数目减少,光度值明显低于SAMR1组(P<0.05);“原络通经”组与百会组小鼠海马组织神经细胞形态与数量较SAMP8空白组明显改善,光度值明显增高(P均<0.05),且“原络通经”组光度值明显高于百会组(P<0.05)。与SAMR1组比较,SAMP8空白组小鼠海马组织中NMDAR蛋白表达量明显降低(P<0.05),海马组织中Aβ_(40)蛋白含量明显增高(P<0.05);与SAMP8空白组比较,“原络通经”组与百会组小鼠海马组织中NMDAR蛋白表达量均明显升高(P均<0.05),海马组织中Aβ_(40)蛋白含量均明显降低(P均<0.05),且“原络通经”组与百会组比较改善更明显(P均<0.05)。结论“原络通经”针刺与百会穴针�Objective It is to observe the effect of“source-collateral acupoint for unblocking the meridian”(SAUM)acupuncture on learning and memory ability of SAMP8 mice,and to explore the mechanism of SAUM acupuncture in promoting the recovery of learning and memory ability of dementia mice.Methods Six SAMR1 mice were selected as the SAMR1 group,which were routinely fed and only scratched every day;another 18 SAMP8 mice were selected and randomly divided into SAMP8 blank group,SAUM group and Baihui group,6 mice in each group,the mice in the SAMP8 blank group were only scratched every day,and the ones in the SAUM group and Baihui group were given corresponding acupuncture interventions.After 30 days of intervention,the learning and memory and spatial exploration abilities of the mice in each group were tested by the Morris water maze test,the changes of neurons in the hippocampus of the mice were observed by HE staining,and the expression of N-methyl-D-aspartic acid receptor(NMDAR)in the hippocampus was detected by Western blot method,and Aβ_(40) protein content in mouse hippocampus was detected by ELISA.Results Compared with the SAMP1 group,the escape latency of the mice in the SAMP8 blank group was significantly prolonged,the residence time in the original platform quadrant was significantly shortened,and the number of crossing the original platform position was significantly reduced,and the differences were statistically significant(all P<0.05).Compared with the SAMP8 blank group,the escape latency of SAMP8 mice in SAUM group and Baihui group were significantly shortened,the residence time in the original platform quadrant was significantly prolonged,and the number of crossing the original platform position was significantly increased,and the improvements were more significantly in SAUM group than those in Baihui group,the differences were statistically significant(all P<0.05).Hippocampal HE staining showed that the shape and structure of the neurons in hippocampus were normal in the SAMR1 group;the neurons in hippo
