摘 要:目的探讨抗帕颗粒与益生菌对帕金森便秘小鼠α-突触核蛋白(α-Syn)在脑肠轴不同部位表达的影响。方法将50只SPF级健康雄性8周龄C57BL/6小鼠随机分为空白组、模型组、益生菌组、抗帕颗粒组、益生菌+抗帕颗粒组,每组10只。除空白组外,其余组小鼠用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)腹腔注射诱导帕金森便秘模型。建模后第8天开始,益生菌组给予酪酸梭菌活菌109个/d灌胃,抗帕颗粒组给予抗帕颗粒40 mg/(kg·d)灌胃,益生菌+抗帕颗粒组给予酪酸梭菌活菌109个/d和抗帕颗粒40 mg/(kg·d)灌胃,模型组和空白组给予0.9%氯化钠溶液1 mL/d灌胃,每日1次,每周按照体重调整1次给药量。连续灌胃4个月后处死各组小鼠,获取结肠、迷走神经腹腔干、大脑黑质纹状体组织,免疫印迹法检测结肠、迷走神经、黑质组织中α-Syn蛋白表达情况,免疫荧光双染检测黑质中酪氨酸羟化酶(TH)、α-Syn表达情况。结果模型组结肠、迷走神经、黑质组织中α-Syn蛋白表达量均明显高于空白组(P均<0.05);益生菌组、抗帕颗粒组、益生菌+抗帕颗粒组结肠、迷走神经、黑质组织中α-Syn蛋白表达量均明显低于模型组(P均<0.05),抗帕颗粒组和益生菌+抗帕颗粒组结肠、迷走神经、黑质组织中α-Syn蛋白表达量均明显低于益生菌组(P均<0.05),且益生菌+抗帕颗粒组结肠、黑质组织中α-Syn蛋白表达量均明显低于抗帕颗粒组(P均<0.05);益生菌+抗帕颗粒组迷走神经、黑质中α-Syn蛋白表达量与空白组比较差异均无统计学意义(P均>0.05)。黑质免疫荧光双染色切片镜下观察,模型组TH染色数量明显少于空白组(P<0.05),益生菌组、抗帕颗粒组、益生菌+抗帕颗粒组均明显多于模型组(P均<0.05);模型组α-Syn染色数量明显多于空白组(P<0.05),益生菌组、抗帕颗粒组、益生菌+抗帕颗粒组均明显少于模型组(P均<0.05)。结论抗帕颗粒和益生菌均可改�Objective It is to investigate the effects of anti-Parkinson’s disease(anti-PD)granules with probiotics onα-synuclein(α-Syn)expression in different parts of brain-gut axis in constipation mice with Parkinson’s disease.Methods Fifty SPF health C57BL/6 male mice aged 8 weeks were randomly divided into blank group,model group,probiotic group,Anti-PD group and probiotic+Anti-PD group,with 10 mice in each group.Except for the blank group,the mice in other groups were given 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)by intraperitoneal injection to establish models of Parkinson’s disease constipation.From the 8th day after modeling,the probiotic group was given 109/d viable Clostridium butyric bacteria by gavage,the Anti-PD group was given 40 mg/(kg·d)Anti-PD granule,and the probiotic+Anti-PD group was given 109/d viable Clostridium butyric bacteria and 40 mg/(kg·d)Anti-PD granule by gavage.The rats in the model group and the blank group were given 1 mL/d 0.9%sodium chloride solution by gavage.All once a day,the dose was adjusted once a week according to body weight.After 4 months of continuous gavage,the mice in each group were sacrificed,and the colon,vagus nerve celiac trunk,and brain nigrostriatum tissues were obtained.The expression ofα-Syn protein in colon,vagus nerve,and substantia nigra tissue were detected by Western blotting,the expression of tyrosine hydroxylase(TH)andα-Syn in substantia nigra was detected by immunofluorescence double staining.Results The expression levels ofα-Syn protein in colon,vagus nerve,and substantia nigra tissue in the model group were significantly higher than those in the blank group(all P<0.05),the expression ofα-Syn protein in colon,vagus nerve,and substantia nigra tissue in the probiotic group,Anti-PD group and probiotic+Anti-PD group was significantly lower than that of the model group(all P<0.05),and the expression ofα-Syn protein in colon,vagus nerve and substantia nigra tissue in the Anti-PD group and probiotic+Anti-PD group were significantly lower lowe
