摘  要:目的:探讨大黄素对内毒素(LPS)诱导的人牙龈成纤维细胞HGF-1炎性因子及活性氧/丝裂原活化蛋白激酶/活化蛋白1(ROS/MAPK/AP-1)通路的影响。方法:将HGF-1细胞分为对照组、LPS组(800μg/L)、大黄素组(60μmol/L)、ROS诱导剂组(ROS诱导剂)、大黄素+ROS诱导剂组。细胞经相应分组处理后,检测细胞活力、炎性因子及RPS/MAPK/AP-1通路蛋白表达水平。结果:与对照组相比,LPS组HGF-1细胞活力降低,炎症因子、丙二醛(MDA)、乳酸脱氢酶(LDH)表达水平、ROS含量、ROS/MAPK/AP-1通路相关蛋白表达水平升高,超氧化物歧化酶(SOD)表达水平降低;与LPS组相比,大黄素组HGF-1细胞活力升高,炎症因子、MDA、LDH表达水平、ROS含量、ROS/MAPK/AP-1通路相关蛋白表达水平降低,SOD表达水平显著升高。结论:大黄素可抑制LPS诱导的牙龈成纤维细胞炎性反应可能是通过抑制ROS/MAPK/AP-1通路实现的。Objective:To investigate the effects of emodin on the expression of inflammatory factors and reactive oxygen species/mitogen-activated protein kinase/activated protein 1(ROS/MAPK/AP-1)pathway in human gingival fibroblast HGF-1 cells induced by LPS.Methods:Human gingival fibroblasts HGF-1 were divided into control group,LPS group(800μg/L),emodin group(60μmol/L),ROS inducer group(ROS inducer),emodin+ROS inducer group.After corresponding treatment,the proliferation,inflammatory factors and ROS/MAPK/AP-1 expression were examined.Results:Compared with the control group,in LPS group the viability of HGF-1 cells was decreased,the expression levels of inflammatory factors,malondialdehyde(MDA)and lactate dehydrogenase(LDH),the content of ROS and the expression levels of ROS/MAPK/AP-1 pathway related proteins were increased,the expression level of superoxide dismutase(SOD)was decreased.Compared with LPS group,in emodin group the activity of HGF-1 cells was increased,the expression levels of inflammatory factors,MDA and LDH,the content of ROS and the expression levels of ROS/MAPK/AP-1 pathway related proteins were decreased,the expression level of SOD was increased.Conclusion:Emodin can inhibit LPS-induced inflammatory response of gingival fibroblasts,which may be achieved by inhibiting ROS/MAPK/AP-1 pathway.
