利多卡因通过miR-146b-5p调控胃癌细胞增殖、迁移、侵袭的分子机制研究

摘  要:目的研究利多卡因是否通过miR-146b-5p调控胃癌细胞的生物学行为。方法胃癌细胞SNU-1以50μmol/L、100μmol/L、200μmol/L利多卡因处理,同时以未处理的细胞为对照组(NC);在SNU-1细胞中转染anti-miR-NC、anti-miR-146b-5p、miR-NC、miR-146b-5p,其中转染miR-NC、miR-146b-5p的细胞以200μmol/L利多卡因处理。利用CCK-8法检测不同处理的SNU-1细胞增殖,流式细胞仪检测细胞凋亡,Transwell检测细胞迁移及侵袭,Western blot检测细胞周期蛋白D1(CyclinD1)、活化的含半胱氨酸的天冬氨酸蛋白水解酶-3(Cleaved Caspase-3)、基质金属蛋白酶2(MMP2)、MMP9蛋白相对表达,实时定量PCR检测miR-146b-5p表达。结果不同浓度利多卡因处理SNU-1细胞后,与NC组比较,50μmol/L利多卡因组、100μmol/L利多卡因组、200μmol/L利多卡因组细胞的增殖能力、迁移数量、侵袭数量、CyclinD1、MMP2、MMP9蛋白和miR-146b-5p的表达量逐渐减弱,凋亡率、Cleaved Caspase-3蛋白表达量逐渐增加,差异均具有统计学意义(P<0.05);转染anti-mi R-146b-5p后SNU-1细胞的miR-146b-5p、CyclinD1、MMP2、MMP9蛋白的表达量、增殖能力、迁移和侵袭数量比转染anti-miR-NC低,Cleaved Caspase-3蛋白表达量、凋亡率比转染anti-miR-NC高,差异均具有统计学意义(P<0.05);miR-146b-5p+200μmol/L利多卡因组SNU-1细胞的CyclinD1、MMP2、MMP9蛋白表达量、增殖能力、迁移和侵袭数量高于mi R-NC+200μmol/L利多卡因组,Cleaved Caspase-3蛋白表达量、凋亡率低于miR-NC+200μmol/L利多卡因组,差异均有统计学意义(P<0.05)。结论利多卡因通过下调胃癌细胞中的miR-146b-5p,抑制细胞增殖、迁移、侵袭,并诱导凋亡。Objective To study whether lidocaine regulates the biological behavior of gastric cancer cells through miR-146b-5p.Methods SNU-1 gastric cancer cells were treated with 50μmol/L,100μmol/L,200μmol/L lidocaine,and untreated cells were used as the control group(NC group).SNU-1 cells were transfected with anti-miR-NC and anti-miR-146b-5p,miR-NC,miR-146b-5p,the cells transfected with miR-NC,miR-146b-5p were treated with200μmol/L lidocaine.CCK-8 method was used to detect the proliferation of SNU-1 cells under different treatments;cell apoptosis was detected by flow cytometry;cell migration and invasion were detected by Transwell;the relative expression of CyclinD1,cleaved-cysteinyl aspartate specific proteinase-3(Cleaved Caspase-3),matrix metalloprotease 2(MMP2),and MMP9 protein were detected by Western blot.The expression of miR-146b-5p was determined by quantitative real-time PCR.Results After treatment of SNU-1 cells with different concentrations of lidocaine,compared with the NC group,the proliferation,the number of migration cells,the number of invasions cells,the expression of CyclinD1,MMP2,MMP9 protein,and miR-146b-5p in the 50μmol/L lidocaine group,100μmol/L lidocaine group,and200μmol/L lidocaine group gradually decreased,and the apoptotic rate and the expression of Cleaved Caspase-3 protein gradually increased,with statistically significant differences(P<0.05).After transfection with anti-miR-146b-5p,the expression level of miR-146b-5p,CyclinD1,MMP2,and MMP9,proliferation capacity,migration and invasion of SNU-1cells were lower than those of anti-miR-NC,and the expression of Cleaved Caspase-3 protein and the rate of apoptosis were higher than those of transfection with anti-miR-NC,all with statistically significant differences(P<0.05).The expression of CyclinD1,MMP2,MMP9 protein,proliferation ability,migration,and invasion numbers of SNU-1 cells in miR-146b-5p+200μmol/L lidocaine group were higher than those in miR-NC+200μmol/L lidocaine group,and Cleaved Caspase-3 protein expression and apoptosis rate
