摘　　要：目的探讨TRIM16在结直肠癌中的表达以及对细胞迁移和侵袭的影响。方法回顾性分析2019年6月至2020年6月于河南科技大学第一附属医院行手术治疗的30例原发性结直肠癌及其正常结直肠组织。检测结直肠癌组织及癌旁正常组织中TRIM16的表达水平;通过在线(https://kmplot.com/)分析结直肠癌组织中TRIM16表达情况与无进展生存期的Kaplan-Meier生存分析;在体外细胞实验观察敲低和过表达TRIM16对结直肠癌细胞迁移、侵袭能力的影响。结果实时荧光定量聚合酶链式反应(RT-PCR)结果显示癌旁组织中TRIM16的表达量明显高于结直肠癌组织,蛋白质印迹结果显示在结直肠癌组织中TRIM16蛋白水平明显低于相应的癌旁组织,免疫组织化学结果同样证明TRIM16在结直肠癌组织相对于癌旁组织明显低表达,差异均有统计学意义(P<0.05)。高表达TRIM16组的患者无进展生存期显著高于TRIM16低表达组,差异有统计学意义(P<0.05)。过表达TRIM16可抑制结直肠癌细胞的迁移和侵袭,敲低则可促进癌细胞的迁移和侵袭。结论TRIM16在结直肠癌中低表达,表达水平与结直肠癌预后相关。推测TRIM16介导的结直肠癌细胞迁移和侵袭抑制是通过其对上皮细胞-间充质转化的抑制作用发挥作用。Objective To investigate the expression of TRIM16 in colorectal cancer and its effect on cell migration and invasion.Methods Thirty patients with primary colorectal cancer and their normal colorectal tissues who underwent surgical treatment in the First Affiliated Hospital of Henan University of Science and Technology from June 2019 to June 2020 were retrospectively analyzed,and detect the expression level of TRIM16 in colorectal cancer tissues and adjacent normal tissues.(https://kmplot.com/)was used to analyze the expression of TRIM16 in colorectal cancer tissues and Kaplan-Meier survival analysis of overall survival and relapse-free survival.The effects of knockdown and overexpression of TRIM16 on the migration and invasion of colorectal cancer cells were observed in vitro.Results Reverse transcription polymerase chain reaction(RT-PCR)indicated that the TRIM16 mRNA levels were significantly decreased in colorectal cancer tissues compared with non-tumor tissues.Consistent with the downregulation of TRIM16 mRNA,Western blotting showed that the protein expression of TRIM16 was significantly decreased in colorectal cancer tissues compared to corresponding non-tumor tissues.IHC consistently showed that TRIM16 expression was decreased in colorectal cancer tissues compared with corresponding non-tumor tissues,the differences were statistically significant(P<0.05).Progression-free survival in patients with high TRIM16 expression was significantly higher than that in patients with low TRIM16 expression,the difference was statistically significant(P<0.05).Overexpression of TRIM16 can inhibit the migration and invasion of colorectal cancer cells,while knockdown can promote the migration and invasion of colorectal cancer cells.Conclusion In this study,it was found that the expression of TRIM16 was low in colorectal cancer,and the expression level was correlated with the prognosis of colorectal cancer.It was speculated that TRIM16 mediated inhibition of migration and invasion of colorectal cancer cells played an important
