摘 要:目的探讨非小细胞肺癌组织中缺氧诱导因子(HIF)-2α、基质金属蛋白酶7(MMP7)的表达及临床意义。方法收集90例非小细胞肺癌患者的非小细胞肺癌组织和癌旁组织,采用免疫组化染色法检测HIF-2α、MMP7的表达情况。比较非小细胞肺癌组织和癌旁组织中HIF-2α和MMP7阳性表达率,比较不同临床特征非小细胞肺癌患者非小细胞肺癌组织中HIF-2α和MMP7阳性表达率,分析HIF-2α与MMP7表达的相关性。结果HIF-2α和MMP7均主要表达于细胞质。非小细胞肺癌组织中HIF-2α阳性表达率为55.56%(50/90),高于癌旁组织的12.22%(11/90),差异有统计学意义(P﹤0.05);非小细胞肺癌组织中MMP7阳性表达率为57.78%(52/90),高于癌旁组织的8.89%(8/90),差异有统计学意义(P﹤0.05)。腺癌、低分化、有淋巴结转移、临床分期为Ⅲ~Ⅳ期非小细胞肺癌患者非小细胞肺癌组织中HIF-2α和MMP7阳性表达率分别明显高于鳞状细胞癌、中高分化、无淋巴结转移、临床分期为Ⅰ~Ⅱ期的患者,差异均有统计学意义(P﹤0.01)。经Spearman等级相关性分析,HIF-2α与MMP7表达呈正相关(r=0.348,P﹤0.05)。结论HIF-2α和MMP7在非小细胞肺癌组织中高表达,其异常表达与肿瘤的浸润和转移有关。Objective To investigate the expression and clinical significance of hypoxia-inducible factor(HIF)-2αand matrix metalloproteinase 7(MMP7)in non-small cell lung cancer tissues.Method The non-small cell lung cancer tissues and adjacent tissues of 90 patients with non-small cell lung cancer were collected,and the expressions of HIF-2αand MMP7 were detected by immunohistochemical staining.The positive rates of HIF-2αand MMP7 in non-small cell lung cancer tissues and adjacent tissues were compared,and the positive rates of HIF-2αand MMP7 in non-small cell lung cancer tissues of patients with different clinical characteristics were also compared,and the correlation between HIF-2αand MMP7 expression was analyzed.Result Both HIF-2αand MMP7 were mainly expressed in the cytoplasm.The pos-itive rate of HIF-2αin non-small cell lung cancer tissues was 55.56%(50/90),which was higher than 12.22%(11/90)in adjacent tissues,and the difference was statistically significant(P<0.05).The positive rate of MMP7 in non-small cell lung cancer tissues was 57.78%(52/90),which was higher than 8.89%(8/90)in adjacent tissues,and the difference was statistically significant(P<0.05).The positive rates of HIF-2αand MMP7 in non-small cell lung cancer tissues of patients with adenocarcinoma,poorly differentiated,lymph node metastasis,and clinical stage III-IV were higher than those with squamous cell carcinoma,moderately-to-well differentiated,no lymph node metastasis,and clinical stage I-II,respective-ly,and the differences were statistically significant(P<0.01).The Spearman rank correlation analysis presented that the ex-pression of HIF-2αwas positively correlated with MMP7(r=0.348,P<0.05).Conclusion HIF-2αand MMP7 are up-regu-lation in non-small cell lung cancer tissues,and their abnormal expression are related to tumor invasion and metastasis.
