摘 要:间充质干细胞(MSCs)是存在于骨髓和大多数结缔组织中的基质细胞群,可分化为骨和软骨等间充质组织。MSCs分化与特定转录因子有关,核心结合因子α1、性别决定区Y框蛋白9、过氧化物酶体增殖物激活受体γ、成肌分化抗原等转录因子在MSCs分化中具有整合作用。转化生长因子-β(TGF-β)家族信号转导由细胞外配体启动,可导致细胞内信号转导介质激活。而TGF-β家族配体TGF-β_(1)、TGF-β_(2)和TGF-β_(3)参与MSCs功能的多个方面,可通过控制关键转录因子的表达和活性调节MSCs的分化功能。因此,深入研究TGF-β家族蛋白在MSCs分化中的作用,有助于未来研究成果向临床的转化。Mesenchymal stem cells(MSCs)are a stromal cell population present in bone marrow and most connective tissues that can differentiate into mesenchymal tissues such as bone and cartilage.The differentiation of MSCs is related to specific transcription factors.Transcription factors such as core binding factorα1,sex-determining region Y-box 9,peroxisome proliferator-activated receptorγ,and myogenic differentiation antigen have integration roles in the differentiation of MSCs.Transforming growth factor-β(TGF-β)family signaling is initiated by extracellular ligands that lead to the activation of intracellular signaling mediators.The TGF-βfamily ligands TGF-β_(1),TGF-β_(2) and TGF-β_(3) are involved in many aspects of MSCs function,and can regulate the differentiation function of MSCs by controlling the expression and activity of key transcription factors.Therefore,in-depth study of the role of TGF-βfamily proteins in the differentiation of MSCs is helpful for the translation of the research results into the clinical practice in the future.
