摘　　要：整合素是一类广泛存在于细胞膜上的跨膜异二聚体,介导细胞与细胞间以及细胞与细胞外基质间的识别和黏附,整合素β_(1)是整合素家族中最大的亚群。整合素β_(1)可以通过结合不同的配体激活不同的下游信号通路,从而介导肿瘤细胞的迁移、侵袭和增殖,抑制肿瘤细胞的凋亡;并且能够通过促进肿瘤血管及淋巴管的生成,促进肿瘤的发生和转移;同时,整合素β_(1)可以通过多种途径增强肿瘤细胞耐药性以及失巢凋亡抗性,影响恶性肿瘤的药物治疗,促进肿瘤转移灶的形成。整合素β_(1)作为恶性肿瘤的潜在治疗靶点,相关作用机制的研究对探索肿瘤的发生、发展以及转移具有重要意义。Integrin is a type of transmembrane heterodimeric protein widely present on the cell membrane,mediating the recognition and adhesion between cells and between cells and extracellular matrix.Integrinβ_(1) is the largest subgroup in the integrin family.A large number of studies have shown that integrinβ_(1) can bind with different ligands to activate different downstream signaling pathways,thus mediating migration,invasion and proliferation of tumor cells and inhibiting apoptosis of tumor cells.And by promoting the tumor angiogenesis and lymphangiogenesis,integrinβ_(1) can promote the occurrence and metastasis of tumors;at the same time,it can enhance drug resistance and anoikis resistance of tumor cells through various ways,influence drug treatment of malignant tumor and promote the formation of tumor metastasis.Integrinβ_(1) is a potential therapeutic target for malignant tumors,and the study of the related mechanisms is of great significance for exploring the genesis,development and metastasis of tumors.
