摘  要:目的探究微小RNA(miR)-937在肝细胞癌组织及癌旁组织的表达,及其表达水平与肝癌患者生存期、临床病理特征与预后的关系。方法选取2017年3月至2019年3月间146例肝癌及其癌旁组织(定义为距离癌灶边缘>2 cm)作为研究对象。采用实时荧光定量聚合酶链反应(PCR)技术检测肝癌组织及癌旁组织中微小miR-937的表达水平;并探究和分析miR-937表达水平与肝癌患者的生存时间、性别、年龄(≥55岁或≤55岁)、甲胎蛋白(AFP,>50 ng/ml或≤50 ng/ml)、组织分化程度、临床分期(Ⅰ~Ⅱ期或Ⅲ~Ⅳ期)的相关性;采用多因素COX分析探究miR-937表达水平与肝癌患者的预后关系。组间比较采用t检验。结果癌旁组织中miR-937表达水平低于肝癌组织中miR-937表达水平(1.04±0.13比2.65±0.49),差异有统计学意义(t=3.159,P<0.05)。临床分期Ⅰ~Ⅱ期患者miR-937表达水平低于Ⅲ~Ⅳ期患者miR-937表达水平(2.43±0.45比2.79±0.34),差异有统计学意义(t=2.873,P<0.05)。miR-937高表达人群的生存时间低于miR-937低表达人群的生存时间(18.7个月比28.6个月),差异有统计学意义(log-rank=4.385,P<0.05)。同时,采用COX回归分析表明肝癌组织中miR-937表达水平能够预测肝癌患者的3年OS率(HR=2.856,P<0.05)。多因素COX分析表明肝癌患者的临床分期(Ⅰ~Ⅱ期比Ⅲ~Ⅳ期)是影响肝癌组织中miR-937表达水平危险因素(P<0.05),miR-937表达水平变化是影响肝癌患者预后的独立影响因素。结论miR-937表达水平在肝癌组织中显著上调,且与患者的临床分期和生存时间明显相关。Objective To investigate the expression of microRNA(miR)-937 in hepatocellular carcinoma(HCC)and adjacent tissues,and its relationship with survival time,clinicopathological features and prognosis of patients with HCC.Methods A total of 146 cases of HCC and its adjacent tissues(defined as>2 cm from the edge of the cancer)collected from March 2017 to March 2019 were selected as the research objects.The expression level of miR-937 in HCC and adjacent tissues was detected by real-time fluorescence quantitative polymerase chain reaction(PCR).The correlation between miR-937 expression level and survival time,gender,age(≥55 years or≤55 years),alpha fetal protein(AFP,>50 ng/ml or≤50 ng/ml),degree of tissue differentiation,clinical stage(stageⅠ-ⅡorⅢ-Ⅳ)in patients with HCC was explored and analyzed.Multivariate Cox analysis was used to explore the relationship between the expression level of miR-937 and the prognosis of patients with HCC.The difference between the groups was compared with t test.Results The expression level of miR-937 in adjacent tissues was significantly lower than that in HCC tissues(1.04±0.13 vs.2.65±0.49,t=3.159,P<0.05).The expression level of miR-937 in patients with clinical stageⅠ-Ⅱwas significantly lower than that in patients with stageⅢ-Ⅳ(2.43±0.45 vs.2.79±0.34,t=2.873,P<0.05).The survival time in patients with high expression of miR-937 was significantly shorter than that in those with low expression of miR-937(18.7 months vs.28.6 months,log-rank=4.385,P<0.05).At the same time,Cox regression analysis showed that the expression level of miR-937 in HCC could predict the 3-year overall survival rate of patients with HCC(HR=2.856,P<0.05).Multivariate Cox analysis showed that the clinical stage(stageⅠ-Ⅱvs.stageⅢ-Ⅳ)of patients with HCC was risk factor affecting the expression level of miR-937 in HCC(P<0.05).Among them,the change of miR-937 expression level was an independent factor affecting the prognosis of patients with HCC.Conclusion The expression level of miR-937
