摘  要:骨肉瘤(OS)是一种常见的原发性恶性骨肿瘤,起源于间叶组织,好发于儿童和青少年,具有耐药,易复发和远处转移等特点。鼠双微体基因2(MDM2)在多种恶性肿瘤中过表达,包括实体瘤、肉瘤和白血病,通过与p53等信号分子的相互作用在肿瘤的发生发展中起着核心作用。近来众多证据表明MDM2基因不仅与骨肉瘤的侵袭、转移等恶性生物学行为有关,对骨肉瘤的治疗及预后也有一定影响。因此,阐明MDM2在骨肉瘤发生发展过程中的作用,对寻求骨肉瘤治疗新靶点具有重要意义,本文就MDM2在骨肉瘤中的研究进展作一综述,旨在为骨肉瘤治疗提供更广阔的思路。Osteosarcoma is a common primary malignant bone tumor originating from mesenchymal tissue.It has a high incidence rate in children and adolescents and exhibits a predilection to drug resistance,recurrence,and distant metastasis.The human homolog of the murine double minute 2(MDM2)oncogene is overexpressed in multiple malignancies,including solid tumors,sarcomas,and leukemia.MDM2 plays a central role in cancer initiationand progression because of its interactions with p53 and other signaling molecules.Recent evidence shows that MDM2 is not only emerging as a regulator of invasion and metastasis but also has a certain influence on the treatment and prognosis of osteosarcoma.Therefore,elucidating the role of MDM2 in the development of osteosarcoma may help us identify novel therapeutic targets.This paper reviews the research progress of MDM2 in osteosarcoma to provide a broader idea for the treatment of osteosarcoma.
