摘  要:目的基于网络药理学探讨“山慈菇-半枝莲”药对治疗肝癌的有效靶点及作用机制。方法通过中药系统药理学数据库分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)数据库检索山慈菇和半枝莲活性成分和相关靶点,在genecards数据库和OMIM数据库获取肝癌相关疾病靶点。运用Cytoscape构建“药物-活性成分-靶点”网络,String数据库构建蛋白相互作用网络(protein-protein interaction,PPI),并通过网络拓扑分析筛选关键成分及核心靶点,最后通过R软件对肝癌靶基因进行GO功能分析和KEGG富集分析。结果本研究筛选出“山慈菇-半枝莲”活性成分30个,肝癌靶点192个,涉及162个信号通路,进一步筛选得出核心化合物7个,核心靶基因6个,GO富集结果显示肝癌靶基因主要参与DNA转录和蛋白降解、炎症反应、免疫抑制以及血管的生成等生理功能。KEGG富集结果显示AGE-RAGE信号通路、乙型肝炎信号通路,IL-17、TNF、PI3K-Akt、NF-kB信号通路等是肝癌靶基因调控的关键信号通路。结论“山慈菇-半枝莲”能通过多种活性成分调节多个肝癌靶基因,发挥多种功能,介导多条信号通路从而发挥抗肝癌作用。Objective To investigate the available targets and mechanism of Pseudobulbus Cremastrae Seu Pleiones-Herba Scutellariae Barbatae(PC-HSB)in treating live cancer based on network pharmacology.Methods The active components and gene targets of PC-HSB were screened by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),targets of live cancer were obtained from GeneCards and Online Mendelian Inheritance in Man(OMIM).Cytoscape was applied to construct the“drug-component-target”network,String Database was used to construct the protein-protein interaction(PPI)network,and the key components and key targets were screened out through network topology analysis.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis of key-targets were performed by R software.Results A total of 30 active components of PC-HSB and 192 liver cancer targets which are involved in 162 signaling pathways were screened out.Subsequently,7 key components and key targets were obtained.GO function enrichment analysis showed that targets of live cancer mainly involve the physiological function of DNA transcription,protein degradation,inflammatory response,immunosuppression and angiogenesis.KEGG pathway enrichment analysis showed that the regulatory mechanism of live cancer targets might be primarily associated with AGE-RAGE,hepatitis B,IL-17,TNF,PI3K-Akt and NF-kB signaling pathway.Conclusion The network pharmacological study intuitively demonstrated that the multi-component,multi-target,multi-functional and multi-pathway pharmacological effects of PC-HSB on liver cancer.
