摘  要:目的 探讨柚皮素对3T3-L1脂肪细胞胰岛素抵抗的作用,和miR-29b过表达在柚皮素与3T3-L1脂肪细胞胰岛素抵抗效应中的关系,进一步研究阐述柚皮素对糖尿病的防治作用机制。方法 利用地塞米松诱导分化成熟的3T3-L1脂肪细胞,并建立胰岛素抵抗模型,用不同浓度柚皮素进行分组干预胰岛素抵抗模型细胞和被miR-29b mimic和inhibitor转染的胰岛素抵抗模型细胞,分别用qRT-PCR和Western blot检测胰岛素信号传导通路中IRS-1、Akt/p-Akt、GLUT4基因和蛋白的表达。结果 柚皮素干预可逆转3T3-L1胰岛素抵抗模型细胞中IRS-1、Akt/p-Akt、GLUT4的基因和蛋白表达的降低;柚皮素通过抑制miR-29b高表达增加3T3-L1胰岛素抵抗模型细胞IRS-1、Akt/p-Akt、GLUT4的基因和蛋白的表达。结论 柚皮素具有改善3T3-L1细胞胰岛素抵抗的作用,其分子机制可能通过抑制miR-29b的高表达影响PI3K/Akt信号通路中相关蛋白的表达。Objective To explore the effect of naringenin on the insulin resistance of 3 T3-L1 adipocytes, and the relationship between the overexpression of miR-29 b and the effects of naringenin on insulin resistance in 3 T3-L1 adipocytes, in order to further illustrate the mechanism by which naringenin acts in the prevention and treatment of T2 DM. Methods Dexamethasone was used to induce differentiated and mature 3 T3-L1 adipocytes, after which an insulin resistance(IR) model was established. Different concentrations of naringenin were used to intervene in IR model cells and IR models cells transfected with miR-29 b mimics and inhibitor. qRT-PCR and Western blot were used to determine the gene and protein expression of IRS-1, Akt/p-Akt and GLUT4. Results Naringenin intervention increased the gene and protein expression of IRS-1, Akt/p-Akt and GLUT4 that was reduced in 3 T3-L1 IR model cells. Naringenin increased 3 T3-L1 IR model cell IRS-1, Akt/p-Akt and GLUT4 gene and protein expression by inhibiting the overexpression of miR-29 b. Conclusions These result show that naringenin can ameliorate insulin resistance in 3 T3-L1 adipocytes, which may improve the expression of related proteins in the PI3 K/Akt signaling pathway by inhibiting the high expression of miR-29 b.
