摘 要:采用化学氧化脱色工艺,对皂素中色素类杂质的共轭键进行破坏,同时保持皂素的基本结构,从而达到粗提皂荚皂素脱色纯化的目的。以H_(2)O_(2)为脱色剂,通过改变脱色剂用量、处理时间、处理温度等因素,获得了粗提皂素脱色纯化的优化工艺。采用分光辐射亮度计对脱色皂素的颜色进行表征,并使用傅里叶红外光谱仪和高效液相色谱仪对皂荚皂素的脱色机理进行探究。脱色纯化处理的优化工艺参数为,脱色剂H_(2)O_(2)用量6%、处理时间2 h、处理温度70℃。粗提皂素经H_(2)O_(2)处理后,颜色由褐色变成米黄色,且色泽自然均匀。脱色皂素水溶液的泡沫高度和半衰期均优于粗提皂素,且具有较低的表面张力,适合在洗护、生物医药等高端领域应用。The chemical oxidation decolorization method was used to destroy the flavonoids in the crude saponin without destroying the basic structure of the saponin.Using H_(2)O_(2) as a decolorizing agent,the optimum decolorization and purification process of saponin from Gleditsia sinensis was obtained by changing the dosage of the decolorizing agent,treatment time,and treatment temperature.The apparent color of decolorized saponin was measured by spectroradiometer,and the decolorization reason of saponin was explored by Fourier transform infrared spectroscopy and high-performance liquid chromatography.The results showed that after the treatment of H_(2)O_(2),the apparent color changed from brown to beige,and the color was uniform.The optimum technological parameters for decolorization and purification of gleditsia saponin by H_(2)O_(2) method were as follows:decolorizer dosage 6%,treatment time 2 h,treatment temperature 70℃.The foam height and half-life of decolorizing saponin aqueous solution are better than those of crude saponin and have lower surface tension,which was suitable for its application in high-level fields such as washing,biological,and medicine.
