超声影像技术对兔肝VX2改良种植瘤生长评估

摘  要:[目的]为探索兔VX2肝癌模型制备的改良种植方法,评估应用超声影像技术对改良种植兔VX2肿瘤的生长情况。[方法]采用从同一个入口并以60°为边界朝3个方向刺破肝组织建立窦道,接种3块肿瘤块在肝内种植法建立兔VX2肝癌模型,种植14 d时采血分离血清肝功能检测、全数字化彩色多普勒超声诊断仪并造影观察肝脏肿瘤回声、血流信号、大小及生长特点、取材进行大体观察、HE染色病理分析。[结果]采用改良的瘤块肝内种植法成功建立兔VX2肝癌模型,成模率为100%。肿瘤种植兔试验组AST、ALT均显著高于对照组。超声诊断仪并造影观察肿瘤种植兔肝脏肿瘤有内部不均质低回声、血流信号、呈现明显的“快进快出”特征。测量出最大、最小直径并计算出肿瘤体积。肉眼观察到肿瘤种植兔肝实质内有鱼肉样灰白色结节状肿瘤组织。HE染色病理观察肿瘤种植兔肝脏有肿瘤的组织学特征。[结论]改良的3个方位瘤块种植法是一种成瘤率高、重复性好、模型性状稳定、可批量建立兔VX2肝癌模型的方法。超声影像技术检查是一种操作简便、能够实时、准确评估兔VX2肝癌的生长特点影像学方法。[Objectives] To explore the improved implantation method for the preparation of rabbit VX2 liver cancer model and evaluate the growth of improved implanted rabbit VX2 tumor by ultrasonic imaging technology.[Methods]The sinuses were established by puncturing the liver tissue in three directions from the same entrance and taking 60° as the boundary,rabbit VX2 liver cancer model was established by inoculating three tumor blocks in the liver,after 14 days of planting,blood was collected and separated for serum liver function detection,full digital color Doppler ultrasound diagnostic instrument and contrast was used to observe the echo,blood flow signal,size and growth characteristics of liver tumor,and the materials were taken for gross observation and the staining pathological analysis.[Results] The rabbit VX2 liver cancer model was successfully established by the improved tumor mass intrahepatic implantation method,and the modeling rate was 100%.AST and ALT in the experimental group were significantly higher than those in the control group.Ultrasound diagnostic instrument and contrast-enhanced observation showed that the liver tumor of tumor implanted rabbits had internal heterogeneous low echo and obvious "fast in and fast out" characteristics of blood flow signal.The maximum and minimum diameters were measured and the tumor volume was calculated.Fish like gray nodular tumor tissue was observed in the liver parenchyma of tumor implanted rabbits.HE staining was used to observe the histological characteristics of tumor implanted rabbit liver.[Conclusions] The improved three orientation tumor block planting method is a method with high tumor formation rate,good repeatability,stable model character and batch establishment of rabbit VX2 liver cancer model.Ultrasound imaging is a simple,real-time and accurate imaging method to evaluate the growth characteristics of rabbit VX2 liver cancer.
