摘  要:目的 探讨结直肠癌组织中环状RNA 0000376(circ_0000376)表达情况及circ_0000376对结直肠癌细胞SW1116生物学行为的影响和潜在机制。方法 实时定量PCR(RT-qPCR)分析21例结直肠癌组织和对应癌旁组织中circ_0000376和微小RNA-330-5p(miR-330-5p)表达水平。分别转染circ_0000376小干扰RNA(si-circ_0000376)、miR-330-5p类似物(mimics)至SW1116细胞,采用细胞计数试剂盒(CCK-8)法和集落形成实验检测SW1116细胞增殖能力,划痕愈合实验检测细胞迁移能力,Transwell实验检测细胞侵袭数量,流式细胞术分析细胞凋亡率。双荧光素酶实验和RT-qPCR验证circ_0000376对miR-330-5p表达的调控作用。结果 结直肠癌组织中circ_0000376表达量明显上调,而miR-330-5p表达量明显下调(P<0.05)。沉默circ_0000376或过表达miR-330-5p均可降低SW1116细胞活力、集落形成数、迁移距离以及细胞侵袭数量(P<0.05),增加细胞凋亡率(P<0.05)。miR-330-5p是circ_0000376的靶基因,沉默circ_0000376显著增加miR-330-5p表达量(P<0.05)。抑制miR-330-5p表达能够减弱沉默circ_0000376对SW1116细胞增殖、迁移、侵袭、凋亡的影响(P<0.05)。结论 结直肠癌组织中circ_0000376呈高表达,沉默circ_0000376通过上调miR-330-5p能够抑制结直肠癌细胞SW1116增殖、迁移和侵袭,并诱导细胞凋亡。Objective To investigate the expression of circular RNA 0000376(circ_0000376)in colorectal cancer tissues,and further to explore the effect of circ_0000376 on the biological behavior of colorectal cancer cells SW1116 and its potential mechanism.Methods The expression levels of circ_0000376 and microRNA-330-5p(miR-330-5p)in 21 cases of colorectal cancer tissues and corresponding adjacent tissues were analyzed using real-time quantitative PCR(RT-qPCR).circ_0000376 small interfering RNA(si-circ_0000376)and miR-330-5p mimics were transfected into SW1116 cells,respectively.Cell counting kit(CCK-8)and colony formation assay were applied to detect the proliferation ability of SW1116 cells,and scratch healing assay was used to calculate cell migration ability,and Transwell assay was selected to measure the number of cell invasion,and flow cytometry was used to analyze cell apoptosis rate.The regulatory effect of circ_0000376 on the expression of miR-330-5p was verified using dual luciferase experiment and RT-qPCR.Results circ_0000376 expression in colorectal cancer tissue was significantly up-regulated(P<0.05),while miR-330-5p expression in colorectal cancer tissue was significantly down-regulated(P<0.05).Silencing circ_0000376 or miR-330-5p overexpression reduced cell viability,colony formation numbers,migration distance and cell invasion number of SW1116 cells(P<0.05),and increased cell apoptosis rate(P<0.05).miR-330-5p is the target gene of circ_0000376,silencing circ_0000376 significantly increased the expression of miR-330-5p(P<0.05).miR-330-5p inhibition could attenuate the effect of silencing circ_0000376 on the proliferation,migration,invasion and apoptosis of SW1116 cells(P<0.05).Conclusion circ_0000376 is highly expressed in colorectal cancer tissues.Silencing circ_0000376 inhibits the proliferation,migration and invasion of colorectal cancer cells SW1116 and induce cell apoptosis by up-regulating miR-330-5p.
