结直肠癌离心性表达Cdc42伴E-cadherin缺失的预后分析

摘　　要：目的探讨Cdc42在结直肠癌中离心性表达伴E-cadherin缺失的临床意义。方法采用免疫荧光和免疫组化法检测Cdc42和E-cadherin在结直肠癌组织中的表达,分析Cdc42表达与临床病理特征和预后的关系;采用实时荧光定量PCR和Western blot法检测上调Cdc42表达后E-cadherin的表达变化。结果45.96%的病例伴Cdc42的离心性表达,与组织浸润、淋巴结转移和远处转移(P<0.05)密切相关,且提示预后更差(P<0.001);Cdc42离心性表达的病例中有73.63%伴E-cadherin表达缺失,E-cadherin表达缺失与组织浸润密切相关(P<0.05),患者预后更差(P<0.001);过表达Cdc42后,E-cadherin表达明显下调。结论Cdc42的离心性表达伴E-cadherin表达缺失,提示结直肠癌的进展和预后不良。Purpose To investigate the clinical significance of the centrifugal expression of Cdc42 in colorectal cancer with E-cadherin deletion.Methods Immunofluorescence and immunohistochemistry were applied to detect the expression distribution of Cdc42 in colorectal cancer tissues and the expression of E-cadherin,and the relationship between its expression and clinicopathological characteristics and prognosis was analyzed.Real-time PCR and Western blot were applied to detect the change of E-cadherin expression after upregulation of Cdc42 expression.Results A total of 45.96%of the cases were accompanied by centrifugal expression of Cdc42,which was closely associated with tissue infiltration,lymph node metastasis and distant metastasis(P<0.05)and suggested a worse prognosis(P<0.001).73.63%of the cases with centrifugal expression of Cdc42 were accompanied by E-cadherin deficiency,and E-cadherin deficiency was closely associated with tumor infiltrating depth(P<0.05)and worse prognosis(P<0.001).E-cadherin expression was significantly downregulated after overexpression of Cdc42.Conclusion The eccentric expression of Cdc42 coupling with the lack of E-cadherin expression indicate the progression and poor prognosis of colorectal cancer.
