摘  要:为了确定提取、分离纯化黑果腺肋花楸(Aronia melanocarpa)中花色苷的最佳工艺,了解黑果腺肋花楸中确切的花色苷单体结构及其抗氧化活性。以黑果腺肋花楸果实为原料,采用单因素试验、响应面分析法探究超声波辅助提取花色苷的最佳工艺条件,并利用高速逆流色谱分离纯化出花色苷单体,并对其抗氧化能力进行测定。结果表明:提取分离纯化黑果腺肋花楸中花色苷的最佳提取条件为超声功率125 W、乙醇体积分数为56%、V(液)∶m(料)=20 mL∶1 g、超声时间4 min,花色苷的最大提取量为6.49 mg·g^(-1);经高速逆流色谱分离纯化制备得到3个组分(矢车菊-3-O-阿拉伯糖苷、矢车菊素-3-O-葡萄糖苷和飞燕草素-3-O-葡萄糖苷),且3个组分对DPPH(1,1-二苯基-2-三硝基苯肼)自由基清除率的能力依次增加。In order to determine the optimal process for extracting,separating and purifying anthocyanins in Aronia melanocarpa,and know the exact monomer structure of anthocyanins in A.melanocarpa and their antioxidant activity.Single factor test and response surface analysis were used to explore the optimal process conditions for ultrasonic-assisted extraction of anthocyanins from A.melanocarpa fruit as the raw material by HSCCC(high-speed countercurrent chromatography),of which the antioxidant capacity was measured.The optimal extraction conditions for the extraction,separation and purification of anthocyanins from A.melanocarpa were ultrasonic power of 125 W,ethanol volume fraction of 56%,V(liquid)∶m(material)of 20 mL∶1 g,ultrasonic wave time at 4 min,and the maximum extraction amount of anthocyanins was 6.49 mg·g^(-1).Three components(cyanidin-3-O-arabinoside,cyanidin-3-O-glucoside and delphinidin-3-O-glucoside)were separated by HSCCC and the ability of the three components to scavenge DPPH(1,1-diphenyl-2-trinitrophenylhydrazine)free radicals increased in turn.
