摘 要:目的:利用生物信息学方法对女性肺腺癌肿瘤组织及正常组织的差异表达基因进行筛选及分析,筛选出女性肺腺癌发生发展的关键基因,并为后续研究提供候选基因。方法:从GEO数据库中下载肺腺癌基因芯片数据集GSE19804、GSE40791、GSE31210、GSE7670、GSE10072、GSE32863、GSE75037,利用在线工具GEO2R筛选出数据集中正常女性肺组织和女性肺腺癌组织的差异表达基因(DEGs),然后利用DAVID数据库对DEGs进行GO和KEGG富集分析,接着利用STRING和Cytoscape软件构建蛋白互相网络(PPI)并筛选出Hub基因,并利用oncomine、UALCAN数据库进行验证,最后使用Kaplan-Meier plotter数据库对筛选出的Hub基因进行生存分析。结果:在7个数据集中共筛选出276个DEGs,其中69个上调,207个下调。共筛选出CDC20、CENPF、KIAA0101、TOP2A、ASPM、TYMS、MCM4、NUSAP1、MELK、UBE2C等10个Hub基因,Kaplan-Meier生存分析显示除ASPM外,其余9个均与女性肺腺癌的总体生存率相关。结论:最终筛选出的10个Hub基因可能与女性肺腺癌的发生发展密切相关,其可能是女性肺腺癌发病机制、预后判断和治疗研究的新靶点。Objective:To screen and analyze the differentially expressed genes of female lung adenocarcinoma and normal lung tissues using bioinformatics method,to screen the key genes related to the carcinogenesis and development of female lung adenocarcinoma,and to provide candidate genes for subsequent studies.Methods:The lung adenocarcinoma microarray datasets GSE19804,GSE40791,GSE31210,GSE7670,GSE10072,GSE32863,and GSE75037 from GEO datasets were downloaded,and the GEO2R online analysis tools were used to screen for the differential expression genes(DEGs)in the female lung adenocarcinoma tissue and the adjacent normal tissue.Enrichment analysis of GO and KEGG pathways were performed by using the DAVID database,and protein interaction network(PPI)web tools STRING and Cytoscape software were constructed and screen out hub genes.The hub genes were selected and combined with the Kaplan-Meier plotter database for further analysis and verification.Oncomine and UALCAN databases were used for verification.Finally,Kaplan-Meier Plotter database was used for survival analysis on the screened hub genes.Results:A total of 276 DEGs were screened out in the 7 data sets,among which 69 were up-regulated and 207 were down-regulated.A total of 10 Hub genes including CDC20,CENPF,KIAA0101,TOP2A,ASPM,TYMS,MCM4,NUSAP1,MELK,and UBE2C were screened.Except ASPM,the remaining 9 genes were related to the overall survival rate of female lung adenocarcinoma in Kaplan-Meier survival analysis.Conclusion:The 10 hub genes selected may be closely related to the occurrence and development of female lung adenocarcinoma,which may serve as new targets for the pathogenesis,clinical diagnosis and treatment of female lung adenocarcinoma.
