摘 要:目的观察黄连素对人肝癌细胞株HepG2增殖及凋亡的影响。方法体外培养人肝癌细胞株HepG2,按不同浓度黄连素(0,1,10,50,100μmol/L)分别作用12,24,36,48 h后,运用CCK-8法检测HepG2细胞增殖,流式细胞术检测HepG2细胞凋亡,Western blot检测Bcl-2、Caspase-3蛋白的表达。结果与0μmol/L黄连素相比,1,10,50,100μmol/L黄连素分别作用于人肝癌细胞株HepG2不同时间后,细胞抑制率随药物浓度及时间的增加而升高(P<0.05)。黄连素以1,10,50,100μmol/L的浓度分别作用48 h后,HepG2细胞凋亡率逐渐升高(F=15.008,P=0.000),且随着黄连素作用浓度的升高,HepG2细胞中Bcl-2蛋白的表达逐渐降低,Caspase-3蛋白的表达逐渐增高,与0μmol/L相比,10,50,100μmol/L黄连素作用后Bcl-2和Caspase-3蛋白表达差异具有统计学意义(P<0.05)。结论黄连素抑制HepG2细胞增殖,诱导HepG2细胞凋亡,其机制可能与caspase级联反应及抗凋亡蛋白Bcl-2减少相关。Objective To observe the effect of berberine on the proliferation and the apoptosis of human hepatocellular carcinoma cell line HepG2.Methods Human hepatocellular carcinoma cells HepG2 were cultured in vitro and treated with 0,1,10,50,100μmol/L berberine,and then the proliferation of HepG2 cells was detected by CCK-8 method,the apoptosis of HepG2 cells was detected by flow cytometry,and the protein expression levels of Bcl-2 and caspsae3 were detected before and after berberine treatment by Western blot.Results Compared with 0μmol/L group,the inhibitory rate of HepG2 cells was increased with the increase of concentration and intervention time after treated with different concentrations of berberine for 12,24,36,48 h(P<0.05).The apoptosis rate of HepG2 cells was gradually increased after treatment with 1,10,50,100μmol/L berberine for 48 h,respectively(F=15.008,P=0.000).With the increase of concentration,the expression of Bcl-2 protein in HepG2 cells was gradually decreased,while the expression of Caspase-3 protein was gradually increased.Compared with 0μmol/L berberine group,Bcl-2 and Caspase-3 proteins showed statistical difference in 10,50,100μmol/L groups(P<0.05).Conclusion Berberine can inhibit the proliferation and induce the apoptosis of HepG2 cells.
