摘 要:目的探究β-榄香烯对宫颈癌细胞增殖与侵袭的作用及机制。方法将对数生长期的HeLa细胞分别用不同浓度的β-榄香烯(0、20、30、40μg/mL)处理后,采用MTT法检测HeLa细胞增殖能力,Transwell检测HeLa细胞的侵袭能力,划痕实验检测HeLa细胞的迁移能力,流式细胞仪检测HeLa细胞凋亡情况,qRT-PCR检测HeLa细胞中NDRG4基因的表达水平、Western blotting检测HeLa细胞中NDRG4蛋白和PI3K/Akt通路相关蛋白的表达水平。结果与β-elemene 0组细胞相比,用不同浓度的β-榄香烯处理后,细胞的增殖能力呈时间和浓度依赖性降低且侵袭细胞数明显减少,划痕闭合率明显降低,细胞凋亡率也呈浓度依赖性增加。不同浓度的β-榄香烯处理组中NDRG4的基因和蛋白表达水平较β-elemene 0组高,而且p-Akt蛋白的表达水平与β-榄香烯的浓度呈依赖性,高浓度组的表达量最低。结论β-榄香烯能通过上调宫颈癌细胞NDRG4的表达水平来抑制细胞增殖、侵袭和转移能力,增加细胞凋亡,从而抑制肿瘤的发展。Objective To investigate the effect and mechanism ofβ-elemene on the proliferation and invasion of cervical cancer cells.Methods Logarithmic growth stage HeLa cells were treated with different concentrations ofβ-elemene(0,20,30 and 40μg/ml).The proliferation ability of HeLa cells was detected by MTT,the invasion ability of HeLa cells by Transwell,the migration ability of HeLa cells by scratch assay and the apoptosis of HeLa cells by flow cytometry.The expression levels of NDRG4 gene in HeLa cells were measured by qRT-PCR,and the expression levels of NDRG4 protein and PI3K/Akt pathway-related protein in HeLa cells were measured by Western blotting.Results Treatment with different concentrations ofβ-elemene resulted in a time-and concentration-dependent decrease in proliferation capacity and a significant decrease in the number of invading cells,a significant decrease in the scratch closure rate and a concentration-dependent increase in the apoptosis rate compared to theβ-elemene 0 group cells.The gene and protein expression levels of NDRG4 were higher in theβ-elemene treated group than in theβ-elemene 0 group at different concentrations,and the expression level of p-Akt protein was dependent on the concentration ofβ-elemene,with the lowest expression in the high concentration group.Conclusionβ-elemene can inhibit cell proliferation,invasion and metastatic ability and increase apoptosis by up-regulating the expression level of NDRG4 in cervical cancer cells,thus inhibiting tumor progression.
