摘 要:目的:探讨环状RNA_0000337(circ_0000337)对胶质瘤细胞增殖、凋亡的影响及其对微小RNA-1299(miR-1299)的调控作用。方法:采用实时荧光定量聚合酶链反应(qRT-PCR)法检测正常脑组织、胶质瘤组织中circ_0000337和miR-1299的表达量;体外培养人胶质瘤细胞U251,si-NC(si-NC组)、si-circ_0000337(si-circ_0000337组)、miR-NC(miR-NC组)、miR-1299 mimics(miR-1299组)、si-circ_0000337和anti-miR-NC(si-circ_0000337+anti-miR-NC组)、si-circ_0000337和anti-miR-1299(si-circ_0000337+anti-miR-1299组)转染入U251细胞;采用CCK-8实验及流式细胞术分别检测细胞增殖及凋亡;双荧光素酶报告基因实验检测circ_0000337与miR-1299的靶向关系。结果:与正常脑组织比较,胶质瘤组织中circ_0000337的表达水平升高(P<0.05),miR-1299的表达水平降低(P<0.05);与si-NC组比较,si-circ_0000337组细胞活力降低(P<0.05),凋亡率升高(P<0.05);与miR-NC组比较,miR-1299组细胞活力降低(P<0.05),凋亡率升高(P<0.05);circ_0000337可靶向调控miR-1299;与si-circ_0000337+anti-miR-NC组比较,si-circ_0000337+anti-miR-1299组细胞活力升高(P<0.05),凋亡率降低(P<0.05)。结论:抑制circ_0000337表达可通过靶向调控miR-1299的表达而抑制胶质瘤细胞增殖及促进细胞凋亡。Objective:To explore the effect of circ_0000337 on the proliferation and apoptosis of glioma cells and its regulatory effect on miR-1299.Methods:The qRT-PCR method was used to detect the expression of circ_0000337 and miR-1299 in normal brain tissue and glioma tissue.In vitro cultured human glioma cells U251,si-NC(si-NC group),si-circ_0000337(si-circ_0000337 group),miR-NC(miR-NC group),miR-1299 mimics(miR-1299 group),si-circ_0000337 and anti-miR-NC(si-circ_0000337+anti-miR-NC group),si-circ_0000337 and anti-miR-1299(si-circ_0000337+anti-miR-1299 group)were transfected into U251 cells.CCK-8 experiment and flow cytometry were used to detect cell proliferation and apoptosis,respectively.The dual luciferase reporter gene experiment was used to detect the targeting relationship between circ_0000337 and miR-1299.Results:Compared with normal brain tissue,the expression level of circ_0000337 in glioma tissue was increased(P<0.05),while the expression level of miR-1299 was decreased(P<0.05).Compared with the si-NC group,the cell viability in the si-circ_0000337 group was decreased(P<0.05),while the apoptosis rate was increased(P<0.05).Compared with the miR-NC group,the cell viability of the miR-1299 group was decreased(P<0.05),while the apoptosis rate was increased(P<0.05).circ_0000337 could target miR-1299.Compared with the si-circ_0000337+anti-miR-NC group,the cell viability in the si-circ_0000337+anti-miR-1299 group was increased(P<0.05),while the apoptosis rate was decreased(P<0.05).Conclusion:Inhibiting the expression of circ_0000337 could inhibit the proliferation of glioma cells and promote cell apoptosis by targeting the expression of miR-1299.
