摘  要:目的探讨三结构域蛋白59(TRIM59)能否通过调控核因子κB(NF-κB)信号通路影响鼻咽癌细胞生物学行为。方法通过癌症基因组图谱(TCGA)数据库预测TRIM59在鼻咽癌和癌旁组织的表达情况;反转录PCR和Western blot法分别检测鼻咽癌和癌旁组织中TRIM59 mRNA和蛋白相对表达量;以人正常鼻黏膜上皮细胞(HNEpC)为对照,采用反转录PCR和Western blot法分别检测HNE1和CNE-2Z鼻咽癌细胞TRIM59 mRNA和蛋白相对表达量;采用小干涉RNA技术下调HNE1细胞TRIM59水平,设置对照组、小干涉RNA阴性对照(si-NC)组、TRIM59小干涉RNA(si-TRIM59)组。噻唑蓝(MTT)法检测各转染组细胞增殖抑制率,TranswellTM小室检测各转染组细胞侵袭和迁移能力,Western blot法检测NF-κB、磷酸化的NF-κB(p-NF-κB)、波形蛋白(vimentin)、存活蛋白(survivin)的蛋白表达,ELISA法检测培养细胞上清液白细胞介素6(IL-6)、IL-8、肿瘤坏死因子α(TNF-α)的含量。结果TRIM59在鼻咽癌组织表达异常增高;与HNEpC细胞相比,TRIM59在HNE1相对表达量显著增加;下调HNE1细胞中TRIM59表达后,显著抑制细胞存活以及细胞的侵袭和迁移;下调TRIM59表达后,HNE1细胞NF-κB、vimentin、survivin mRNA以及p-NF-κB、vimentin、survivin蛋白表达均受到抑制,IL-6、IL-8、TNF-α含量显著降低。结论下调TRIM59能够抑制HNE1细胞NF-κB信号通路和肿瘤炎症相关因子进而抑制细胞侵袭和迁移。Objective To investigate whether tripartite motif containing 59(TRIM59)influences the biological behavior of nasopharyngeal carcinoma cells by regulating the nuclear factor kB(NF-kB)signaling pathway.Methods TCGA database was used to predict the expression of TRIM59 in nasopharyngeal carcinoma and adjacent tissues.Reverse transcription PCR and Western blot were used to detect the relative expressions of TRIM59 mRNA and protein in nasopharyngeal carcinoma and paracancerous tissues.With human normal nasal mucosal epithelial cells(HNEpCs)as a control,reverse transcription PCR and Western blot analysis were used to detect the relative expression of TRIM59 mRNA and protein in HNEl and CNE-2Z nasopharyngeal carcinoma cells.Small interference RNA technology was used to down-regulate the level of TRIM59 in HNEl cells,while a control group,small interference RNA negative control(si-NC)group and TRIM59 small interference RNA(si-TRIM59)group were set up.MTT assay was used to detect the cell proliferation inhibition rate of each transfection group.Transwell™chamber was used to detect cell invasion and migration ability of each transfection group.Western blot analysis was employed to detect NF-kB and phosphorylated NF-kB(p-NF-kB),vimentin,survivin protein expression.ELISA was adopted to detect interleukin 6(IL-6),IL-8,tumor necrosis factor a(TNF-a)in the supernatant of cultured cells.Results The expression of TRIM59 increased abnormally in nasopharyngeal carcinoma tissues.Compared with HNEpC cells,the relative expression of TRIM59 in HNEl significantly increased.After down-regulating the expression of TRIM59 in HNEl cells,cell survival and cell invasion and migration were significantly inhibited.Down-regulating the expression of TRIM59 inhibited HNE1 cells p-NF-kB,vimentin,survivin protein expression and significantly reduced the levels of IL-6,IL-8,and TNF-a.Conclusion Down-regulation of TRIM59 blocks the NF-kB signaling pathway and tumor inflammation-related factors in HNEl cells,thereby inhibiting cell invasion and migra
