摘 要:目的分析双特异性磷酸酶1(Dual specificity phosphatase 1,DUSP1)在宫颈鳞状细胞癌(简称宫颈鳞癌)中的表达水平及其作用。方法收集2017年1月-2019年5月在新疆医科大学附属肿瘤医院妇科收治的73例Ⅰ-Ⅳ期宫颈鳞癌患者的肿瘤组织、癌旁组织及其临床病理资料。采用qRT PCR方法检测肿瘤组织及其癌旁组织中DUSP1的表达水平,分析其与临床病理参数的相关性。采用过表达DUSP1的慢病毒进行转染,设为C33a实验组和SiHa实验组;用携带无义序列的慢病毒进行转染,设为C33a对照组和SiHa对照组,共4组。用CCK-8和Transwell实验分别检测细胞增殖、侵袭及迁移能力;Western blot检测C33a实验组和对照组中的MAPK信号通路相关蛋白磷酸化水平。结果宫颈鳞癌组织中DUSP1的表达水平低于癌旁组织,无淋巴结转移患者DUSP1表达水平高于有淋巴结转移患者(P<0.05)。与C33a对照组和SiHa对照组比较,C33a实验组和SiHa实验组宫颈鳞癌细胞DUSP1表达水平显著增加(P<0.01),细胞增殖(P<0.05)、迁移(P<0.01)、侵袭(P<0.01)能力降低。与C33a对照组比较,C33a实验组宫颈鳞癌细胞p-p38、p-JNK、p-ERK的蛋白磷酸化表达水平降低(P<0.05)。结论DUSP1的过表达水平与宫颈鳞癌细胞增殖、迁移、侵袭能力相关,可能是潜在的抑癌基因,有望成为临床治疗宫颈鳞癌的新型靶标。Objective To analyze the expression level and function of dual specificity phosphatase 1(DUSP1)in cervi⁃cal squamous cell carcinoma.Methods The tumor tissues and adjacent tissues of 73 patients with stage I-IV cervical squamous cell carcinoma were admitted to the hospital from March 2017 to May 2019 were selected.The expression level of DUSP1 in tumor tissue and its adjacent tissue were detected by qRT-PCR,and its relationship with clinical in⁃dicators was analyzed.The overexpressed DUSP1 and control sequences were transfected into C33a and SiHa cells by lentivirus transfection,and cells were divided into C33a experimental group,SiHa experimental group,C33a control group and SiHa control group.Cell proliferation and invasion and migration activity were detected by CCK-8 and Tran⁃swell,respectively.Protein phosphorylation related with MAPK signaling in C33a experimental group and C33a con⁃trol group were detected by Western blot.Results The expression level of DUSP1 in cervical cancer tissues was signifi⁃cantly lower than that in adjacent tissues and related to lymph node metastasis(P<0.05).Compared with C33a control group and SiHa control group,the expression level of DUSP1 in C33a and SiHa groups were increased(P<0.01),and the cell proliferation(P<0.05),migration(P<0.01),invasion(P<0.01)were significantly decreased.Compared with C33a control group,the protein expression levels of p-p38,p-JNK and p-ERK in C33a experimental group were de⁃creased(P<0.05).Conclusion The expression level of DUSP1 was negatively correlated with the proliferation,migra⁃tion and invasion of cervical squamous cell carcinoma.DUSP1 may be a potential tumor suppressor gene and may be used as a new clinical diagnostic indicator of cervical squamous cell carcinoma.Thus,the mechanism of DUSP1 affect⁃ed cervical squamous carcinoma cells pathogenesis involved in inhibition of protein phosphorylation of MAPK signal⁃ing pathway.
