摘 要:以红花注射液的副产物为原料,通过大孔树脂柱吸附技术和超滤膜分离工艺相结合的方法,优选红花黄色素的最佳纯化和精制工艺。通过单因素考察大孔树脂型号、吸附时间、洗脱醇度、药材-树脂比、超滤膜除杂孔径、超滤膜浓缩孔径对纯化工艺的影响,并通过中试验证优选的纯化工艺。结果表明,最佳的纯化工艺条件为:采用D101大孔树脂,吸附5h,40%乙醇四倍体积洗脱,洗脱液采用3K超滤膜精制,然后再采用1K超滤膜浓缩,浓缩液喷雾干燥即得红花黄色素粉末。经三批中试结果验证,色价均在160以上,回收率接近50%,产率达到了10%以上,说明该工艺适用于红花注射液副产物开发红花黄色素的大批量生产。Using the by-product of safflower injection as raw material,through the method of combining macroporous resin column adsorption technology andultrafiltration membrane separation process,the best purification and refining process of safflower yellow pigment is optimized.The effect of macroporous resin model,adsorption time,elution alcohol degree,medicinal material-resin ratio,pore size ofultrafiltration membrane and concentration pore size ofultrafiltration membrane on the purification process was investigated by single factor,and the optimal purification process was verified by pilot test.The results showed that the best purification process conditions were:D101 macroporous resin,adsorption for 5 hours,four times the volume of 40% ethanol for elution,the eluate was refined with a 3K ultrafiltration membrane,and then concentrated with a 1Kultrafiltration membrane.The concentrate was spray-dried to obtain carthamus yellow powder.The results of three batches of pilot tests showed that the color price was above 160,the recovery rate was close to 50%,and the yield reached more than 10%,indicating that the process is suitable for the large-scale production of carthamus yellow developed by the by-product of safflower injection.
