摘　　要：目的研究UBE2Q1靶向PI3K/AKT通路对胃癌细胞凋亡、增殖的影响。方法采用生物信息学方法和免疫组织化学方法分析胃癌组织、癌旁组织中UBE2Q1的表达;采用RT-qPCR方法筛选UBE2Q1高表达胃癌细胞株;Annexin V-APC/PI双染色法和流式细胞术检测UBE2Q1对胃癌细胞凋亡、增殖的影响;Western blot法检测UBE2Q1对PI3K/AKT信号通路的影响。结果UBE2Q1在胃癌组织和胃癌细胞系中明显表达上调,UBE2Q1在MKN-28细胞株中的表达最高;抑制UBE2Q1表达可以促进细胞凋亡并影响细胞周期;UBE2Q1低表达组的p-PI3K/PI3K和p-AKT/AKT水平明显下降。结论UBE2Q1通过PI3K/AKT信号通路促进胃癌细胞增殖和抑制细胞凋亡。Objective To study the effect of UBE2Q1 targeting PI3K/AKT pathway on apoptosis and proliferation of gastric cancer cells.Methods The expression of UBE2Q1 in gastric cancer and adjacent tissues was analyzed by bioinformatics and immunohistochemistry;The gastric cancer cell line with high expression of UBE2Q1 was screened by RT-qPCR;Annexin V-APC/PI double staining and flow cytometry were used to detect the effect of UBE2Q1 on apoptosis and proliferation of gastric cancer cells.The effect of UBE2Q1 on PI3K/AKT signaling pathway was detected by Western blot.Results The expression of UBE2Q1 was significantly up-regulated in gastric cancer tissues and gastric cancer cell lines,and the expression of UBE2Q1was the highest in MKN-28 cell line.Inhibition of UBE2Q1 expression can promote apoptosis and affect cell cycle.The levels of p-PI3K/PI3K and p-AKT/AKT in UBE2Q1 low expression group decreased significantly.Conclusion UBE2Q1 promotes the proliferation and inhibits apoptosis of gastric cancer cells through PI3K/AKT signaling pathway.
