摘　　要：目的探讨下调Burkitt淋巴瘤Raji细胞NCAPG表达对细胞增殖和凋亡的影响及其机制。方法应用shR-NCAPG慢病毒载体转染Raji细胞,qRT-PCR及Western blot验证细胞中NCAPG表达是否下调。应用CCK-8方法检测shR-NCAPG对Raji细胞增殖的影响,流式细胞术Annexin V-PE/7AAD双染检测细胞凋亡。Western blot法检测shR-NCAPG转染前后Bcl-2和Bax的表达变化。结果shR-NCAPG转染Raji细胞后,NCAPG在基因及蛋白水平表达减少,明显抑制Raji细胞的增殖,增加细胞早期凋亡比例,抑制Bcl-2表达,上调Bax表达。结论下调NCAPG表达抑制Burkitt淋巴瘤细胞Raji增殖诱导其凋亡,与Bcl-2家族蛋白相关。Objective To explore the effect of downregulating NCAPG expression on the proliferation and apoptosis of Burkitt lymphoma Raji cells.Methods shR-NCAPG lentiviral vector was transfected into Raji cells in which the expression of NCAPG was determined by qRT-PCR and Western blot.The proliferation of Raji cells was detected by CCK-8 kit.The apoptosis of Raji cells was stained by Annexin V-PE/7 AAD.Western blot was used to detect the expression of Bcl-2 and Bax in Raji cells.Results The expression levels of NCAPG protein and m RNA was decreased after transfection of shR-NCAPG into Raji cells.ShR-NCAPG significantly inhibited the proliferation of Raji cells,increased Raji cells proportion of early apoptosis and Bax expression level,but inhibited Bcl-2 expression.Conclusion Downregulated NCAPG expression inhibits the proliferation of Raji and induces apoptosis,which is related to Bcl-2 family proteins.
