摘  要:目的:探讨臭氧应激对大鼠气道黏膜细胞群落和黏蛋白表达谱的影响.方法:SD大鼠随机分为正常对照组、臭氧应激组.扫描电镜观察各组大鼠气管上皮细胞群落的变化.实时荧光定量PCR技术检测Muc5ac、Muc5b mRNA表达量,通过PAS染色测定黏液储备量.结果:正常对照组大鼠气管被纤毛细胞覆盖,随着臭氧持续应激,纤毛细胞脱落加剧,Clara细胞数量减少,杯状细胞增多,未在纤毛细胞脱落处观察到杯状细胞.RT-PCR结果显示臭氧应激组大鼠气道内Muc5ac、Muc5b mRNA的表达量均高于正常对照组,与臭氧应激时间成正相关.结论:臭氧应激大鼠气管细胞群落及黏蛋白基因表达谱发生改变,二者呈正相关关系.Objective Explore the relationship between changes in airway epithelial cells communities and mucin expression profiles in asthma rats models.Methods SD rats were randomly divided into Normal control group and group ozone stress.Compared the changes in cell populations in the lungs of animals at different times of ozone stress with the control group by scanning electron microscopy.By applying real-time PCR experiments we measured Muc5ac,Muc5b mRNA levels.Using PAS staining we measured the total amount of mucin.Results Tracheal epithelium is almost covered by ciliated cells completely.While ozone stress aggravates,increased shedding of ciliated cells was seen,and the number of Clara cells gradually reduced,while the number of goblet cells increased.But goblet cells were not observed in the ciliated cell shedding place.RT-PCR results:both rat airway Muc5ac and Muc5b mRNA and total protein expression level upregulated in ozone stress groups compared to the normal control group.Conclusion We conclude that:mucin gene expression profile changes should be attributed to changes in cell populations in asthma rats models.
