摘 要:目的 研究姜黄素介导丝裂原活化蛋白激酶(MAPK)信号通路对非小细胞肺癌细胞Bcl-2、Bcl-xl及Bax蛋白表达的影响。方法 对非小细胞肺癌A549细胞实施体外培养、分组,并用10μmol/L、20μmol/L、40μmol/L姜黄素处理24 h后,借助显微镜对A549细胞形态结构进行观察;通过MTT法、流式细胞仪对A549细胞增殖抑制率、凋亡率进行测定,经免疫细胞化学法测定MAPK的阳性表达,免疫印迹法测定Bcl-2、Bcl-xl、Bax蛋白表达。结果 姜黄素组的非小细胞肺癌A549细胞增殖抑制率高于对照组(P <0.05);姜黄素组的非小细胞肺癌A549细胞凋亡率高于对照组(P <0.05);姜黄素组的非小细胞肺癌A549细胞内MAPK阳性表达率低于对照组(P <0.05)。姜黄素组A549细胞内Bcl-2、Bcl-xl蛋白浓度低于对照组,Bax蛋白浓度高于对照组(P <0.05)。结论 姜黄素能介导MAPK信号通路,通过下调Bcl-2、Bcl-xl,上调Bax蛋白表达,抑制非小细胞肺癌细胞增殖,促进癌细胞凋亡。Objective To study the effect of curcumin-mediated mitogen-activated protein kinase( MAPK) signaling pathway on the expression of Bcl-2,Bcl-xl and Bax proteins in non-small cell lung cancer cells. Methods A549 cells of non-small cell lung cancer were cultured in vitro,grouped,and treated with 10 μmol/L,20 μmol/L,40 μmol/L curcumin for 24 hours,and then the morphology and structure of A549 cells were observed with a microscope. The A549 cell proliferation inhibition rate and apoptosis rate were determined by MTT method and flow cytometry,and the positive expression of MAPK was determined by immunocytochemistry. Western blotting was used to detect the expression of Bcl-2,Bcl-xl and Bax protein.Results The non-small cell lung cancer A549 cell proliferation inhibition rate in the curcumin group was higher than that in the control group( P < 0. 05). The apoptosis rate of non-small cell lung cancer A549 cells in the curcumin group was higher than that in the control group( P < 0. 05). The positive expression rate of MAPK in non-small cell lung cancer A549 cells in the curcumin group was lower than that in the control group( P < 0. 05). The concentration of Bcl-2 and Bcl-xl protein in A549 cells in the curcumin group was lower than that of the control group,and the concentration of Bax protein was higher than those in the control group( P < 0. 05). Conclusion Curcumin can mediate the MAPK signal pathway,and cancer cell apoptosis is promoted by down-regulating Bcl-2,Bcl-xl,up-regulating the expression of Bax protein,and inhibiting the proliferation of non-small cell lung cancer cells.
