摘 要:目的探讨半枝莲多糖对宫颈癌荷瘤小鼠的抑瘤作用及机制。方法荷瘤小鼠随机分为模型组、LPA组、半枝莲组和半枝莲+LPA组,各15只。绘制肿瘤生长曲线;称取肿瘤质量,计算抑瘤率;检测肿瘤组织细胞凋亡和细胞周期;HE染色观察肿瘤组织病理学情况;免疫组织化学染色观察B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)阳性表达率;Western blot法检侧肿瘤组织雷帕霉素靶蛋白(mTOR)、p-mTOR、P70核糖体蛋白S6激酶(P70S6K)、p-P70S6K蛋白表达。结果半枝莲多糖可增加肿瘤细胞凋亡,抑制肿瘤生长,降低mTOR/P70S6K通路相关蛋白表达,而半枝莲联合使用LPA则可减弱半枝莲多糖的抑瘤作用。结论半枝莲多糖可能通过调控mTOR/P70S6K通路对宫颈癌荷瘤小鼠发挥抑瘤作用。Objective To investigate the antitumor effect and mechanism of scutellaria barbata polysaccharide on cervical cancer tumor-bearing mice.Methods Tumor-bearing mice were randomly divided into model group,LPA group,scutellaria barbata group and scutellaria barbata+LPA group,with 15 mice in each group.Tumor growth curve was drawed,tumor mass was weighed and inhibition rates were calculated,tumor cell apoptosis and cell cycle were detected,HE staining was used to observe tumor histopathology,immunohistochemical staining was used to observe B cell lymphoma-2(Bcl-2)and Bcl-2 associated X protein(Bax)positive expression rates,Western blot was used to detect the protein expression of mammalian target of rapamycin(mTOR),p-mTOR,P70 ribosomal protein S6 kinase(P70S6K)and p-P70S6K in the lateral tumor tissue.Results Scutellaria barbata polysaccharide can increase tumor cell apoptosis,inhibit tumor growth,and reduce the expression of mTOR/P70S6K pathway-related proteins,while the combined use of LPA can attenuate the tumor-inhibitory effect of scutellaria barbata polysaccharide.Conclusion Scutellaria barbata polysaccharide may exert an inhibitory effect on cervical cancer tumor-bearing mice by regulating the mTOR/P70S6K pathway.
