摘 要:目的本研究基于生物信息学分析,探寻肝细胞癌(hepatocellular carcinoma,HCC)发生发展的枢纽基因,为HCC患者的预后提供潜在的生物标志物。方法从GEO数据库下载两个肝细胞癌芯片数据(GSE14520和GSE46408),通过GEO2R筛选HCC组织和正常肝组织之间的差异表达基因(DEGs)。利用STRING数据库构建DEGs的蛋白质互作(PPI)网络,并用Cytoscape软件的CytoHubba插件计算PPI中各基因的连接度,筛选出枢纽基因。通过利用ONCOMINE、GEPIA2、cBioPortal、GeneMANIA、DAVID6.8和TIMER数据库对筛选出的枢纽基因进行转录、翻译、生存情况和肿瘤免疫浸润相关性等方面的验证,并分析其是否具有作为HCC预后生物标志物的潜力。结果筛选出3个枢纽基因(CCNB1、ASPM和AURKA),且均在HCC中高表达。生存分析表明这3个基因均与HCC患者的总体生存率和无病生存率显著相关。肿瘤免疫浸润分析结果进一步提示CCNB1、ASPM和AURKA可能是HCC潜在的预后标志物。结论CCNB1、ASPM和AURKA可能是HCC发生发展的枢纽基因,具有成为HCC预后标志物的潜力。Objective This study was based on bioinformatics analysis to explore key genes in the development of hepatocellular carcinoma(HCC)and to provide potential biomarkers for the prognosis of HCC patients.Methods Two HCC microarray data(GSE14520 and GSE46408)were downloaded from the GEO database,and differentially expressed genes(DEGs)between HCC tissues and normal liver tissues were screened by GEO2R.The protein interaction(PPI)network of DEGs was constructed using the STRING database,and the CytoHubba plug-in of Cytoscape software was used to calculate the connectivity of each gene in the PPI and screen the key genes.The screened key genes were validated for transcription,translation,survival and tumor immune infiltration correlation by using ONCOMINE,GEPIA2,cBioPortal,GeneMANIA,DAVID6.8 and TIMER databases,and analyzed for their potential as prognostic biomarkers for HCC.Results Three key genes(CCNB1,ASPM and AURKA)were screened,and all were highly expressed in HCC.Survival analysis showed that all 3 genes were significantly associated with overall survival and disease-free survival of HCC patients.The results of tumor immune infiltration analysis further suggested that CCNB1,ASPM and AURKA may be potential prognostic markers for HCC.Conclusion CCNB1,ASPM and AURKA may be key genes in the development of HCC and have the potential to become prognostic markers for HCC.
