摘 要:目的探讨罗伊氏FLRE5K1乳酸杆菌发酵乳改善顺铂致小鼠肝损伤及肠道菌群紊乱的效果。方法将24只雄性C57BL/6小鼠随机正常对照组(C组)、L.reuteri FLRE5K1组(FLRE5K1组)、顺铂组(DDP组)和顺铂+L.reuteri FLRE5K1组(DDP+FLRE5K1组)。FLRE5K1组每天给予100μL菌浓度为109cfu/mL的L.reuteri FLRE5K1发酵乳灌胃,DDP组隔天给予腹腔注射25mg/kg顺铂,DDP+FLRE5K1组小鼠同时给予腹腔隔天注射顺铂和每天L.reuteri FLRE5K1发酵乳菌灌胃。20 d后,检测各组小鼠血清中ALT、AST、TB和DB的水平,取小鼠肝右叶边缘肝组织,苏木精—伊红染色后观察肝组织损伤情况,采集小鼠新鲜粪便,对其进行16SrRNA高通量测序。结果DDP组血清ALT及AST水平分别为109.51 U/L和514.19 U/L,显著高于C组和FLRE5K1组;DDP+FLRE5K1组小鼠血清ALT和AST水平分别为83.07U/L和405.31 U/L,明显低于DDP组。肝脏病理切片可见DDP+FLRE5K1组小鼠肝脏较DDP组小鼠肝细胞水肿减轻,未见气球样变细胞。与DDP组小鼠相比,DDP+FLRE5K1组小鼠肠道微生物的丰富度和多样性增加,厚壁菌门和变形菌门丰富度升高,拟杆菌门丰富度降低。DDP+FLRE5K1组小鼠粪便中Barnesiella属、Oscillibacter属、Prevotella属和Alistipes属丰富度较DDP组升高,而Clostridium_XlVa属、Helicobacter属、Acetatifactor属和Alloprevotella属丰富度较DDP组下降。结论摄入L.reuteri FLRE5K1能减轻顺铂诱导的肝损伤并对肠道菌群紊乱具有一定良好的改善作用。Objective To explore the effect of Lactobacillus reuteri FLRE5K1 supplementation on cisplatin-induced liver injury and intestinal flora disorder in mice.Methods 24 male C57BL/6 mice were randomly divided into normal control group(C group),L.reuteri FLRE5K1 group(FLRE5K1 group),cisplatin group(DDP group)and cisplatin+L.reuteri FLRE5K1 group(DDP+FLRE5K1 group).FLRE5K1 group was given 100μl of L.reuteri FLRE5K1 milk with a concentration of 109cfu/mL every day,DDP group was given intraperitoneal injection of 25mg/kg cisplatin once every two days,and DDP+FLRE5K1 group mice were given intraperitoneal injection of cisplatin and L.reuteri FLRE5K1 fermented milk bacteria at the same time.After 20 days,the levels of ALT,AST,TB and DB were measured.After hematoxylin eosin staining,the damage of liver tissue at the edge of the right lobe of mice was observed.At the same time,fresh feces of mice in each group were taken for 16SrRNA high-throughput sequencing.Results The levels of serum ALT and AST in DDP group were 109.51 U/L and 514.19 U/L,respectively,which were significantly higher than those in C group and FLRE5K1 group;The levels of serum ALT and AST in DDP+FLRE5K1 group were 83.07U/L and 405.31 U/L respectively,which were significantly lower than those in DDP group.The liver pathological section showed that the liver edema of mice in DDP+FLRE5K1 group was less than that in DDP group,and there were no balloon like cells.Compared with DDP group,the abundance and diversity of intestinal microorganisms in DDP+FLRE5K1 group increased,the richness of Firmicutes and Proteus increased,and the richness of Bacteroidetes decreased.The abundance of barnesiella,oscillibacter,Prevotella and alistipes in the feces of DDP+FLRE5K1 group was higher than that of DDP group,while the richness of Clostridium_XlVa,Helicobacter,acetatifactor and alloprevotella decreased compared with DDP group.Conclusion Supplementation of L.reuteri FLRE5K1 can effectively reduce the liver injury and intestinal flora disorder induced by cisplatin.
