脱水穿心莲内酯对子宫内膜癌细胞增殖和凋亡的影响

摘　　要：目的研究脱水穿心莲内酯对子宫内膜癌细胞增殖和凋亡的影响。方法体外培养人子宫内膜癌细胞Ishikawa,加入浓度为40、160、320μmol/L的脱水穿心莲内酯作用24 h,MTT法检测细胞增殖率,流式细胞仪检测细胞凋亡,Western blot测caspase-3蛋白和Bcl-2蛋白表达。结果与对照组相比,脱水穿心莲内酯可显著抑制子宫内膜癌细胞增殖(P<0.05),抑制作用具有浓度依赖性;同时,不同浓度脱水穿心莲内酯处理细胞24 h后,细胞存活率均明显降低(P<0.05),作用具有药物浓度依赖性。Western blot结果表明,脱水穿心莲内酯可导致细胞内caspase-3表达水平上调和Bcl-2表达水平下调(P<0.05)。结论脱水穿心莲内酯对人子宫内膜癌细胞的增殖具有明显的抑制作用,同时可诱导子宫内膜癌细胞凋亡,可能是通过影响caspase-3和Bcl-2表达水平而发挥作用的。Objective To study the effects of dehydratedandrographolide on proliferation and apoptosis of endometrial cancer cells.Methods Human endometrial cancer cell line Ishikawa was cultured in vitro and treated with dehydratedandrographolide at concentrations of 40,160 and 320μmol/L for 24 h.The cell proliferation rate was detected by MTT assay,apoptosis was detected by flow cytometry,and the expression of caspase-3 protein and Bcl-2 protein were detected by Western blot.Results Compared with the control group,dehydroandrographolide could significantly inhibit the proliferation of endometrial cancer cells(P<0.05);The inhibition was concentration dependent.At the same time,after treated with different concentrations of dehydratedandrographolide for 24 hours,the cell survival rate decreased significantly(P<0.05);The effect is drug concentration dependent.The results of Western blot showed that dehydratedandrographolide could up regulate the expression of caspase-3 and down regulate the expression of Bcl-2(P<0.05).Conclusion Dehydroandrographolide can significantly inhibit the proliferation of human endometrial cancer cells and induce apoptosis of endometrial cancer cells,which may play a role by affecting the expression levels of caspase-3 and Bcl-2.
