摘  要:目的设计并合成一个用于过氧化氢(H_(2)O_(2))可视化检测的新型小分子生物发光探针(HPBP)。方法在萤火虫萤光素的6’位羟基引入特异性识别基团芳基硼酸酯,构建H_(2)O_(2)小分子探针HPBP。通过探针与底物反应后光学信号强度的变化,对H_(2)O_(2)检测的特异性和灵敏度进行考察。结果成功制备了HPBP,该探针对低浓度H_(2)O_(2)(0.25~5μmol·L^(-1))的响应具有良好的浓度依赖性,且在H_(2)O_(2)浓度低于其他物质20~300倍时,发光强度仍为其他物质的2~5倍。结论制备的HPBP探针检测限低,特异性强,为研究H_(2)O_(2)在疾病发生发展过程中的机制和作用提供了有力工具。OBJECTIVE To develop a novel bioluminescent probe(HPBP)for visual detection of hydrogen peroxide(H_(2)O_(2)).METHODS The probe HPBP was synthesized by introducing an aryl boronate group to the 6’-hydroxy group of D-luciferin.The specificity and sensitivity of probe HPBP were evaluated by monitoring the fluctuation of luminescent intensity after triggered by H_(2)O_(2).RESULTS The bioluminescent probe HPBP was designed for the chemoselective visualization of low concentration H_(2)O_(2)(0.25-5μmol·L^(-1)).There is a good linearity between the bioluminescence intensities and H_(2)O_(2)levels.Moreover,the luminescent intensity generated from the reaction of the probe with H_(2)O_(2)was 2-5 times higher than that with other substances,where H_(2)O_(2)concentration was 20-300 times lower.CONCLUSION The probe HPBP with low limit of detection and strong specificity is developed,providing a powerful tool for mechanism study of hydrogen peroxide in the occurrence and development of diseases.
