摘　　要：目的分析SERPINE1在肝癌组织中的表达水平,探讨其对肝癌细胞增殖的影响及其调控机制。方法利用GEPIA网站分析SERPINE1在肝癌及癌旁组织中的表达情况。荧光定量PCR检测五株肝癌细胞中SERPINE1的表达,选择SERPINE1低表达的细胞转染SERPINE1过表达载体,SERPINE1高表达的细胞转染SERPINE1干扰片段,细胞转染48 h后,CCK-8检测细胞增殖情况;双荧光素酶以及荧光定量PCR检测miR-660对SERPINE1的调控作用,CCK-8进一步验证SERPINE1是否介导了miR-660对肝癌细胞增殖的调控。结果TCGA数据库分析结果显示,SERPINE1在肝癌组织中的表达水平显著降低(P<0.05)。CCK-8结果显示,与空载体组相比,过表达SERPINE1显著抑制了肝癌细胞的增殖(P<0.05);双荧光素酶结果显示,miR-660能够结合SERPINE1,并且上调miR-660抑制了SERPINE1的表达(P<0.05)。此外,与NC mimic组相比,过表达miR-660显著促进了细胞增殖(P<0.05),而与miR-660-mimic+Vector组相比,上调miR-660-mimic+SERPINE1显著抑制了细胞增殖(P<0.05)。结论SERPINE1能够抑制肝癌细胞的增殖,并介导miR-660对肝癌细胞增殖的调控。Objective To analyze the expression level of SERPINE1 in hepatocellular carcinoma tissue and explore its effect on the proliferation of hepatocellular carcinoma cells and its regulated mechanism.Methods The expression of SERPINE1 in hepatocellular carcinoma and adjacent tissues was analyzed by using GEPIA website.The expression of SERPINE1 in 5 hepatoma cell lines was determined by real-time PCR.The cells with low expression of SERPINE1 was transfected with SERPINE1 overexpression vector,and the cells with high expression of SERPINE1 was transfected with siRNA-SERPINE1.CCK-8 was used to detect the cell proliferation at 48h after transfection.The regulated effect of mir-660 on SERPINE1 was detected by double luciferase and real-time PCR.CCK-8 was further verified whether SERPINE1 mediated miR-660 on the regulation of cell proliferation.Results TCGA database analysis showed that the expression level of SERPINE1 in hepatocellular carcinoma tissues decreased significantly(P<0.05).CCK-8 results showed that overexpression of SERPINE1 significantly inhibited the proliferation of hepatoma cells(P<0.05).Double-luciferase results showed that miR-660 could bind SERPINE1(P<0.05).Furthermore,compared with NC mimic group,overexpression of miR-660 significantly promoted cell proliferation(P<0.05),while compared with miR-660-mimic+Vector group the up-regulation of miR-660-mimic+SERPINE1 significantly inhibited cell proliferation(P<0.05).Conclusion SERPINE1 can inhibit the proliferation of hepatoma cells,and involved in miR-660 mediate regulation of the proliferation of liver cancer cells.
