摘  要:目的探索脐带间充质干细胞(umbilical cord mesenchymal stem cells,UCMSCs)培养上清液对食管鳞癌(esophageal squamous cell carcinoma,ESCC)细胞系KYSE450成球的作用。方法收集UCMSCs培养上清液并制备冻干粉,在对KYSE450进行成球培养过程中加入含有不同浓度上清液冻干粉的类器官培养基(organoid medium,OM),利用高内涵成像系统观察各浓度组KYSE450的成球情况。结果KYSE450细胞接种24 h后(Day 1)有少量细胞聚集生长,随后各组细胞球直径逐渐增大;培养至第6天(Day 5),OM组、添加上清液冻干粉浓度30μg/ml组及3μg/ml组出现部分细胞球崩解;第7天(Day 6),3000μg/ml组及300μg/ml组细胞团仍能保持完整的细胞球形态,其他组均有大量细胞球崩解。结论UCMSCs上清液可以促进KYSE450成球。Objective To explore the effect of the supernatant of umbilical cord mesenchymal stem cells(UCMSCS)on the spheroidization of esophageal squamous cell carcinoma(ESCC)cell line KYSE450.Methods The culture supernatant of UCMSCs was collected and made into lyophilized powder;organoid medium(OM)containing different concentrations of supernatant lyophilized powder was added during the spherogenesis of KYSE450 and the spherogenesis of KYSE450 in each group was observed using high-inclusion imaging system.Results After 24 hours of cell seeding(Day 1),a small number of cells grew in aggregates,followed by a gradual increase in the diameter of cell spheres in each group;on the 6th day(Day 5),some cell spheres disintegrated in OM group,30μg/ml group and 3μg/ml group;on the 7th day(Day 6),the cell spheres in 3000μg/ml and 300μg/ml groups remained intact,while a large number of cell spheres disintegrated in all other groups.Conclusion UCMSCs supernatant can promote sphere-formation KYSE450.
