福建省龙岩地区蜱携带的新型弹状病毒全基因序列分析及流行病学调查

摘  要:采集寄生于野猪、牛体表的蜱988只,其中台湾革蜱59只、微小牛蜱12只、台湾血蜱103只、豪猪血蜱214只和微小扇头蜱600只。根据蜱种、宿主、采集地点选取388只代表性蜱进行宏转录组测序分析,获得一个弹状病毒新种的基因组信息,采用逆转录-聚合酶链反应(RT-PCR)与cDNA末端快速扩增技术(RACE)获取了该病毒全基因组序列,命名为龙岩蜱病毒(Longyan tick virus, LYTV)。通过对该病毒的全基因组序列特征分析发现,LYTV具有典型的Ledantevirus病毒属病毒的基因组结构特征。同源性分析表明,LYTV与最近亲缘病毒的核苷酸序列同源性为47.1%~59.1%,RNA依赖性RNA聚合酶L的氨基酸序列同源性为47.0%~56.1%,核蛋白N的氨基酸序列同源性为39.0%~54.6%。通过系统进化分析发现,LYTV与Nkolbisson virus(NKOV)亲缘关系最近,并与A亚群中的Yongjia tick virus 2(YjTV-2)、NKOV、Nishimuro virus(NISV)、Barur virus(BARV)、Fukuoka virus(FUKV)形成独立的分支,表明LYTV为弹状病毒科Ledantevirus病毒属A亚群中的一个新种。流行病学调查结果显示,LYTV在采集蜱样品中的总阳性率为1.7%(6/135),其中豪猪血蜱和台湾血蜱的阳性率分别为2.7%(5/18),1.0%(1/8),推测血蜱可能是LYTV主要传播媒介或贮存宿主。本研究在福建省龙岩地区的蜱中发现了一种新型弹状病毒,为新发蜱传病的防控提供了基础数据。In order to understand the diversity of viruses carried by ticks in Longyan, Fujian Province, 988 ticks were collected from wild boars and cattle, including D.taiwanensis(59),B.microplus(12),H.taiwanensis(103),H.hystricis Supino(214), and R.microplus(600).A total of 388 representative ticks were randomly selected according to tick species, host, and location of collection for meta-transcriptomic analysis.Several contigs were mapped to rhabdovirus.The complete genomic sequence of the virus was obtained by reverse transcription polymerase chain reaction(RT-PCR) and rapid amplification of cDNA ends(RACE),named Longyan Tick virus(LYTV).LYTV had the typical genomic structure of the genus Ledantevirus.Homology analysis showed that the nucleotide sequence homology between LYTV and its closest relatives ranged from 47.1% to 59.1%,the amino acid homology of L protein ranged from 47.0% to 56.1%,and the amino acid homology of N protein ranged from 39.0% to 54.6%.Phylogenetic analysis showed that LYTV was closely related to NKOV and formed an independent clade with YjTV-2,NKOV,NISV,BARV and FUKV of subgroup A,indicating that LYTV may be a novel species in subgroup A of genus Ledantevirus.Epidemiological investigation showed that the positive rate of LYTV in the tick samples was 1.7%(6/135).The positive rate was 2.7%(5/18)in H.hystricis Supino and1.0%(1/8)in H.taiwanensis,suggesting that Haemaphysalis species is the main vector of transmission and reservoir of LYTV.The presented study found a novel rhabdovirus and its prevalence in ticks from Longyan of Fujian Province,which may provide basic information for the prevention and control of its potential outbreak in the future.
