摘 要:试验探究了利用杏鲍菇残渣制备D-氨基葡萄糖盐酸盐的工艺。考察了料液比、反应时间、反应温度、盐酸浓度4个单因素变量对D-氨基葡萄糖盐酸盐得率的影响,在此基础上进行了L9(34)正交试验以确定最佳工艺,按照最佳工艺制备了D-氨基葡萄糖盐酸盐,并测定样品的氯离子含量和比旋光度,利用纸层析法和红外光谱对样品进行初步鉴定。结果显示,利用杏鲍菇残渣制备D-氨基葡萄糖盐酸盐的理想工艺为:反应时间3.5 h、料液比1∶6、反应温度100℃、盐酸浓度30%,D-氨基葡萄糖盐酸盐的得率可达5.23%;经测定样品中氯离子含量为18.14%,层析分析和红外光谱分析表明该方法制备的D-氨基葡萄糖盐酸盐的纯度较高。工艺的确定为利用杏鲍菇残渣制备D-氨基葡萄糖盐酸盐提供了参考。The preparation of D-glucosamine hydrochloride from Pleurotus eryngii residue was investigated.The effects of solid-liquid ratio,reaction time,reaction temperature and concentration of hydrochloric acid on the yield of D-glucosamine hydrochloride were investigated.On this basis,L9(34)orthogonal test was carried out to determine the optimal process.D-glucosamine hydrochloride was prepared according to the optimal process,and the chlorine ion content and specific rotation of the sample were determined.The samples were identified by paper chromatography and infrared spectroscopy.The results showed that the optimal preparation conditions of D-glucosamine hydrochloride from P.eryngii residue were as follows:reaction time 3.5 h,solid-liquid ratio 1:6,reaction temperature 100℃,hydrochloric acid concentration 30%,the yield of D-glucosamine hydrochloride could reach 5.23%.The content of chlorine ion in the sample was 18.14%.The chromatographic analysis and spectral analysis showed that the purity of D-glucosamine hydrochloride prepared by this method was high.This process provided reference for preparing D-glucosamine hydrochloride from P.eryngii residue.
