摘  要:目的:研究蛇床子素及其植物半乳糖脂肝靶向脂质体对人肝癌细胞HepG-2的影响。方法:不同浓度蛇床子素及脂质体处理HepG-2细胞后,采用CCK-8法检测细胞增殖抑制率;流式细胞仪检测细胞摄取、细胞周期变化;Annexin V/PI双染法检测细胞凋亡;Western Blot检测p53表达水平。结果:蛇床子素及其脂质体对肝癌细胞HepG-2均具有明显的抑制作用。与游离蛇床子素相比,脂质体明显增强细胞吸收,显著增强对HepG-2细胞的增殖抑制、诱导凋亡、细胞周期G1期阻滞作用,明显下调p53的表达。结论:蛇床子素植物半乳糖脂肝靶向脂质体比游离蛇床子素具有更强的抗肝癌HepG-2细胞的作用。Objective:To study the inhibitory effect of osthole and osthole-loaded plant galactolipid liver-targeted liposome(OGL) on HepG-2 cells.Methods:The CCK-8 method was used to test the proliferation inhibition rate of HepG-2 cells after treated with osthole(40, 20, 10, 5, 2.50 μg/ml, OGL(40, 20, 10, 5, 2.50 μg/ml) in corresponding concentration for different time(24, 48 h). Flow cytometry was used to detect the cell adoption and the cell cycle of HepG-2 cells after treated with osthole(10 μg/ml), OGL(10 μg/ml) for 24 h. The cell apoptosis was detected by Annexin V/PI. The p53 was detected by Western blot. Results:Osthole, OGL obviously inhibited the activation of HepG-2 cells in a time and concentration dependent manner, the effects of OGL was more stronger. Compared to osthole, the OGL significantly increased the adoption of drug, the proliferation inhibition, the induction of cell apoptosis, the G1 arrest, and the expression of p53. Conclusion:OGL has the more stronger effect against HepG-2 cells than that of osthole.
