摘  要:为了解非洲猪瘟病毒(ASFV)多基因家族成员MGF360-10L基因及其编码蛋白的理化性质和结构特征,本研究在NCBI数据库中搜集到68株含有MGF360-10L序列的ASFV毒株,对这些序列进行比较并构建系统发育进化树;分析MGF360-10L基因及其编码蛋白的理化性质,预测蛋白二、三级结构;利用重组质粒验证在线网站预测的亚细胞定位结果。结果显示,MGF360-10L基因在Ⅱ型毒株中长度相同,基因同源性较高,与其他基因型毒株存在一定差异;理化性质分析结果显示,MGF360-10L蛋白不稳定系数为34.07,二级结构主要是ɑ-螺旋,说明该蛋白较稳定;在249~271 aa处存在跨膜区,不含核定位序列;间接免疫荧光试验结果也证实MGF360-10L蛋白定位在细胞质中。本研究探讨了MGF360-10L基因及其编码蛋白的结构特征,为了解其结构与功能间的关系提供了数据。In order to understand the physical and chemical properties and structural characteristicsof the MGF360-10L gene and its encoded protein,a member of the African swine fever virus(ASFV)multi-gene family,this study collected 68 ASFV strains containing MGF360-10L sequence in the NCBI database.These nucleotide sequences were compared and a phylogenetic tree was constructed;physical and chemical properties of the MGF360-10L encoded protein were analyzed;the secondary and tertiary structure of the protein were predicted;then recombinant plasmid was constructed to verify the subcellular localization which predicted by the online website.The results showed these lengths of MGF360-10L in genotypeⅡstrains were same with high homology,while certain differences existed between different genotype strains;the analysis of physical and chemical properties showed that the protein instability index of MGF360-10L was 34.07,with its secondary structure mainlyɑ-helix,indicating the protein is relatively stable;a transmembrane region was predicted at 249—271 amino acid while no nuclear localization sequence exist.The indirect immunofluorescence assay also confirmed that MGF360-10L protein is localized in the cytoplasm.This study explored the structural characteristics of MGF360-10L gene and its encoded protein,and provide datato understand its structure and function.
