摘  要:根际和根内微生物群落对植物的属性和功能具有重要的影响。湿地植物与微生物协同作用,驱动湿地生态系统的物质循环过程,去除污染物,净化水体。本研究运用高通量测序对不同污染梯度下水葱(Scirpus validus)根际、根内细菌群落的结构和多样性进行了分析,研究了环境因子对植物微生物组多样性的影响。结果表明:水葱根际和根内细菌群落在门水平上的组成相似性达69.23%,主要由厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、放线菌门(Actinobacteriota)等组成。根际与根内细菌群落的优势属组成差异性达76.92%。根内的优势属主要为微小杆菌属(Exiguobacterium)、红球菌属(Rhodococcus)、硫杆菌属(Thiobacillus)、芽孢杆菌属(Bacillus)以及norank_f Steroidobacteraceae属。db-RDA分析结果表明,环境因子对根际细菌多样性的影响较大,总解释率达51.93%,而对根内细菌多样性影响较小,总解释率为39.99%。pH、NO_(3)^(-)、NH_(4)^(+)、总氮对根际细菌群落结构的形成起主要作用,而根内细菌群落结构主要受pH和NH_(4)^(+)的影响。分子生态网络分析表明:根际和根内细菌群落结构的复杂性具有显著差异,根际细菌群落的物种数、多样性和网络复杂性较高,生态位分化更为明显。Rhizosphere and endophytic microbial communities have important effects on the properties and functions of plants.Plants and microorganisms work together to drive the material cycle process of wetland ecosystem,remove pollutants and purify water.High-throughput sequencing was used to analyze the structure and diversity of the rhizosphere and endophytic bacterial communities of Scirpus validus along different pollution gradients,and to study the effects of environmental factors on structure and diversity of plant microbiome.The results showed that the composition of rhizosphere and endophytic bacterial communities of S.validus was similar at the phylum level(69.23%),which was mainly composed of Firmicutes,Proteobacteria,Actinobacteria and so on.The composition of dominant genera in the rhizosphere and endophytic bacterial communities was quite different(76.92%).The dominant genera in endophytic community were mainly Exiguobacterium,Rhodococcus,Thiobacillus,Bacillus and norank_F steroidobacteraceae.The results of db-RDA analysis showed that the environmental factors had a great impact on rhizosphere bacterial diversity,with a total explanation ratio of 51.93%,while had little impact on endophytic bacterial diversity,with a total explanation rate of 39.99%.pH,NO_(3)^(-),NH_(4)^(+)and total nitrogen(TN)played a major role in the formation of rhizosphere bacterial community structure,while the endophytic bacterial community structure was mainly affected by pH and NH_(4)^(+).The results from molecular ecological network analysis showed that there were significant differences in the complexity between rhizosphere and endophytic bacterial community structure.The species,diversity and network complexity of rhizosphere bacterial community were higher,and the niche differentiation of bacterial community was more obvious.
