摘 要:目的探讨长链非编码RNA lnc-SLC2A12-10∶1沉默对人胃癌细胞系AGS增殖、迁移和侵袭的影响。方法用基因干扰技术构建lnc-SLC2A12-10∶1沉默的AGS细胞株。RT-qPCR验证lnc-SLC2A12-10∶1的干扰效率;CCK-8法检测AGS细胞增殖;划痕和Transwell小室法检测AGS细胞迁移和侵袭。软件预测lnc-SLC2A12-10∶1靶向微小RNA(miRNA)并RT-qPCR验证。结果Lnc-SLC2A12-10∶1 shRNA能有效干扰lnc-SLC2A12-10∶1的表达,lnc-SLC2A12-10∶1沉默可抑制胃癌细胞增殖(P<0.05)。沉默lnc-SLC2A12-10∶1还抑制AGS细胞的迁移和侵袭(P<0.05)。经验证,lnc-SLC2A12-10∶1靶向的miRNA可能为hsa-miR-105-5p和hsa-miR-150-3p。结论沉默lnc-SLC2A12-10∶1可显著抑制AGS细胞增殖、迁移和侵袭能力。Objective To investigate the effect of long non-coding RNA(lncRNA)lnc-SLC2A12-10∶1 silencing on the proliferation,migration and invasion of gastric cancer cell line AGS.Methods Gene interference technology was used to construct the lnc-SLC2A12-10∶1 silenced gastric cancer cell strain AGS,and RT-qPCR was used to verify the interference efficiency by lnc-SLC2A12-10∶1 silencing.The proliferation of AGS cell was examined by CCK-8 method.The cell migration and invasion were evaluated by wound healing and Transwell assay.Bioinformatics prediction software was applied to predict lnc-SLC2A12-10∶1 targeting miRNA and to verify them with RT-qPCR.Results Lnc-SLC2A12-10∶1 shRNA effectively silenced expression of lnc-SLC2A12-10∶1,and lnc-SLC2A12-10∶1 silencing inhibited the proliferation of gastric cancer cells(P<0.05).Lnc-SLC2A12-10∶1 silencing also inhibited the migration and invasion of AGS cells(P<0.05).It had been demonstrated that lnc-SLC2A12-10∶1 targeting miRNAs might be hsa-miR-105-5p and hsa-miR-150-3p.Conclusions Lnc-SLC2A12-10∶1 silencing can significantly inhibit the proliferation,migration and invasion of AGS cells.
