摘  要:以黑籽南瓜(Cucurbita ficifolia Bouché)作为原材料,采用超声波辅助法提取黑籽南瓜黄酮类化合物,利用响应面法优化提取工艺,高效液相色谱-质谱联用技术(HPLC-MS/MS)对黑籽南瓜黄酮类化合物进行成分分析并评价其抗炎活性。结果表明,当料液比1:20 g/mL、乙醇体积分数70%、超声时间50 min、超声功率250 W时,黑籽南瓜黄酮类化合物的提取率最高,为3.531%;HPLC-MS/MS结果表明,黑籽南瓜黄酮类化合物主要有6种,分别为金丝桃苷、芦丁、大波斯菊苷(芹菜素-7-O-葡萄糖苷)、槲皮素、木犀草素-7-O-葡萄糖甙、甲基条叶蓟素(3-甲氧基蓟黄素),通过对各峰面积积分可知槲皮素含量最高,占黄酮类化合物总含量的45%;黑籽南瓜黄酮类化合物能够有效抑制脂多糖(LPS)诱导的小鼠单核巨噬细胞(RAW264.7)产生炎症因子一氧化氮(NO)和前列腺素E_(2)(PGE_(2)),证明其具有抗炎活性。Taking Cucurbita ficifolia Bouchéas raw material,the flavonoids of Cucurbita ficifolia Bouchéwere extracted by ultrasonic assisted extraction.Taking the extraction rate of flavonoids as the index,the optimal extraction process was determined by single factor experiment and response surface optimization.High performance liquid chromatography-mass spectrometry(HPLC-MS/MS)was used to analyze the flavonoids composition,its anti-inflammatory activity was evaluated.The results showed that when the solid-liquid ratio was 1:20(g/mL),the volume fraction of ethanol was 70%,the ultrasonic time was 50 min,and the ultrasonic power was 250 W,the extraction rate of flavonoids from Cucurbita ficifolia Bouchéwas the highest,which was 3.531%.The results of HPLC-MS/MS showed that there were 6 kinds of flavonoids in Cucurbita ficifolia Bouché,which were r utin,quercetin,luteolin-7-o-glucoside,hypericin(apigenin-7-o-glucoside),hypericin and methyl striped thistle(3-methoxythistle flavin).According to the area integral of each peak,the content of quercetin is the highest,accounting for 45%of the total content of flavonoids.Cucurbita ficifolia Bouchéflavone extract can effectively inhibit the production of inflammatory factors nitric oxide(NO)and prostaglandin E_(2)(PGE_(2))by mouse monocyte macrophages(RAW264.7)induced by lipopolysaccharide(LPS),proving its anti-inflammatory activity.
